摘 要:目的研究桂枝茯苓对宫颈癌侵袭和转移的影响,并初步探讨其作用机制。方法选用Transwell侵袭系统,观察桂枝茯苓对宫颈癌细胞侵袭转移的影响;用逆转录聚合酶链反应(RT-PCR)、Western blot法检测桂枝茯苓对宫颈癌细胞MMP-2、MMP-9 mRNA及蛋白表达的影响。结果桂枝茯苓100 mg/ml组、50 mg/ml组穿过的细胞数分别为9.00±1.58、9.60±2.07,较空白对照组的40.00±3.81明显减少(P<0.05)。桂枝茯苓能够明显降低宫颈癌细胞MMP-2、MMP-9 mRNA及蛋白表达水平(P均<0.05)。结论桂枝茯苓能有效抑制宫颈癌侵袭和转移,其作用机制可能与降低宫颈癌细胞运动能力,抑制MMP-2、MMP-9的表达水平有关。[ Objective] To investigate the effect of guizhi-fuling on the invasion and metastasis of cervical cancer in vitro. [ Methods] The effect of guizhi-fuling on invasive ability of HeLa cells was observed with a transweU cell culture chamber. The mRNA expression of MMP-2 and MMP-9 in HeLa cells was detected by RT-PCR. And the protein expression was detected by Western blotting. [ Results] In the transweU cell culture chambers, invading HeLa cells in both the groups of 100 mg/ml, 50 mg/ml guizhi-fuling (9.00 ± 1.58 and 9.60 ±2.07) were significantly lower than those in the control group (40.00 ±3.81 ), P 〈0.05. The levels of mRNA and protein of MMP-2 and MMP-9 were decreased by resveratrol (P 〈 0. 05 ). [ Conclusion ] Guizhi-fuling can effectually inhibit the migration and invasion of HeLa cells in vitro. The mechanism may be related to down-regulating the expression of MMP-2 and MMP-9.
