摘  要:目的研究P物质(SP)对大鼠垂体前叶(AP)培养细胞内Ca2+的影响,进而阐述SP的作用机制。方法原代培养成年雌性S-D大鼠的AP细胞48h后,加入不同浓度的SP孵育30min,采用Fura-2/AM作为细胞内钙离子的荧光探针负载培养的AP细胞,结合阳离子测定系统检测AP细胞[Ca2+]i。结果孵育时间为30min时,SP使AP细胞内Ca2+的水平增加,且呈剂量依赖性。结论Fura-2/AM结合阳离子测定系统可以精确的反映细胞内游离钙离子浓度,SP通过SPR产生的生物学效应可通过第二信使Ca2+来完成,在SP对生殖轴(下丘脑-垂体前叶-卵巢/睾丸)调控的理论中进一步证明了第二信使转导途径的作用。Objective This thesis was to determine whether SP had an effect on the free Ca^2+ concentration in primary cultured AP cells of the rat. It further indicated that SP induced the action mechanism of physiology effect. Methods We chose adult female S - D rat AP culture 48h and added different concentration of SP for 30min. After incubating, Fura - 2/AM was loaded as calcium fluorescent probe. The changes of Ca^2 + in AP cells were observed by cation meaturement system. Results When time course is 30 min, SP increased the free Ca^2+ concentration in the AP cells in a concentration dependent manner. Conclusions The system can accurately reflect the concentration of free Ca^2+ in AP cells. Biological responses of SP following activation of SPR are mediated by the second messenger Ca^2+. The information transforming effect of the second messenger is further confirmed in the theory of SP controlling the reproducing axis.
