摘 要:目的:初步探讨治疗乳腺癌的作用机制是否与调节机体免疫功能有关。方法:采用4T1乳腺癌小鼠为实验模型,口服中药"乳癌术后方",观察其抑瘤及抗肺转移作用,并对荷瘤小鼠T细胞增殖功能、T细胞表型及细胞因子IL-10、IL-12、IFNγ-等免疫指标进行检测。结果:实验显示"乳癌术后方"对荷瘤小鼠瘤体积抑制率及肺转移抑制率分别为42.70%、13.14%,与模型组相比能显著抑制肿瘤生长(P<0.01);中药与CTX同时使用还能将CTX的抑瘤率再提高13%(P<0.01),肺转移抑制率再提高近10%。与模型组相比,"乳腺癌术后方"能明显上调荷瘤小鼠T细胞和B细胞活性,下调巨噬细胞百分比,明显增强T淋巴细胞增殖能力(P<0.01),并能抑制荷瘤小鼠淋巴细胞IL-10的生成,增强IFNγ-及IL-12的产生,使之更接近正常小鼠水平。结论:"乳癌术后方"的抗肿瘤作用可能是通过减轻荷瘤宿主的免疫抑制、增强机体抗肿瘤免疫反应来实现的。Objective:To explore whether the mechanism for the anti-breast cancer effects of " Ruaishuhou herbs" was related with the regulation of the immunological system of the host. Methods. We used 4TI mice as highly metastatic breast cancer model. 4TI mice were administered " Ruaishuhou herbs" intragastrically. The anti-tumor and anti-lung metastasis effects of " Ruaishuhou herbs" were observed. The proliferation and phenotypes of T lymphocytes and cytokine levels such as IL-10, IL-12, IFN-γ were tested. Re- sults:Intragastrical administration of "Ruaishuhou herbs" significantly inhibited the tumor volume by 42.70% and lung metastasis by 13. ,14%. The difference was significant compared with the model group ( P 〈0.01 ). CTX inhibited the tumor volume by 55.52% and lung metastasis by 87.13%. " Ruaishuhou herbs" combined with CTX treatment further inhibited the tumor volume by 68.74% and lung metastasis by 97.05%. The difference between combined therapy group and single CTX treatment group was significant ( P 〈 0.01 ). Compared with model group, " Ruaishuhou herbs" up-regulated the activity of T and B lymphocytes, down-regulated the percentage of macrophages, and enhanced the proliferation of T lymphocytes in tumor-bearing mice (P 〈 0. 01 ). " Ruaishuhou herbs" decreased the levels of IL-10 and increased the level of IFN-~ and IL-12. The levels of IL-10, IFN-γ, and IL-12 were near to those in normal mice after " Ruaishuhou herbs" treatment. Conclusion:" Ruaishuhou herbs" may exert anti-tumor effects through relieving the immunosuppression and enhancing anti-tumor immune reaction of tumor-bearing hosts.
