摘  要:目的:探讨左旋咪唑(LMS)对大鼠实验性自身免疫性脑脊髓炎(EAE)脊髓IL-10、IL-12/23P40mRNA表达的影响。方法:用GPSCH和CFA混匀乳剂免疫Wistar大鼠制作EAE模型,分别在免疫前(LMS1组)、免疫同时(LMS2组)、免疫后(LMS3组)给LMS10mg·kg-1。比较各组行为学变化,用RT-PCR测脊髓IL-10和IL-12/23P40mRNA的表达。结果:LMS1组和LMS2组行为学表现加重(P<0.01,P<0.05)。LMS3组出现EAE复发。与EAE组比,LMS1组、LMS2组和LMS3组脊髓IL-12/23P40mRNA表达不同程度升高,IL-10mRNA表达不同程度降低。结论:LMS可升高脊髓IL-12/23P40mRNA表达,降低IL-10mRNA表达,提示LMS对EAE的促发作用可能与Th1/Th2失衡有关。Aim: To study the effects of levamisole (LMS) given in different stages on the expressions of IL-10,IL-12/23 P40 mRNA in spinal cords of rats with experimental autoimmune encephalomyelitis(EAE). Methods: EAE of Wistar Rats was induced by immunized with guinea pig spinal cord homogenate (GPSCH) and complete Freund's adjuvant (CFA) emulsion. LMS was given before or simultaneously or after immunization at a dose of 10 mg·kg^-1. The effect of levamisole on the incidence, latency period, clinical score of EAE rats were compared by methods of quantization. Meanwhile the expressions of IL-10, IL-12/23 P40 mRNA in spinal cords of different groups were compared by reverse transcription-polymerase chain reaction (RTPCR). Results: LMS given before immunization (LMS 1) and LMS given at the same time of immunization (LMS2) could aggravate EAE. LMS given discontinuously after immunization (LMS3) caused relapse in EAE rats. Spinal cords from rats of LMS2 and LMS1 showed increased level of IL-12/23 P40 mRNA, while levels of IL-10 mRNA was decreased as compared to EAE. There was more expression of IL-12/23 P40 mRNA in the relapse phase of EAE (LMS3) compared with acute phase of EAE (EAE), but expression of IL-10 mRNA was no statistical difference between these two phases. Conclusion: LMS could increase the expression of IL-12/23 P40 mRNA and decrease the expression of IL-10 mRNA in spinal cords. The shiftting of Th1/Th2 cytokine balance which caused by LMS might be result in promoting EAE.
