摘  要:目的探讨甲状腺相关眼病(thyroid-associated ophthal mopathy,TAO)患者眼眶前脂肪细胞分化前后相关基因表达的改变,进一步验证脂肪细胞分化在TAO发病中的重要作用。方法培养TAO患者(14例)眼眶前脂肪细胞,并诱导其向脂肪细胞分化,采用RT-PCR法检测分化前后脂肪细胞分化转录因子前脂肪细胞因子-1(Pref-1)、过氧化物酶体增殖体激活受体γ(PPARγ)、脂肪细胞分泌因子瘦素(Leptin)、脂联素(Adiponec-tin)以及共同抗原促甲状腺激素受体(TSHR)5个基因mRNA表达的改变。结果RT-PCR法测的TAO眼眶前脂肪细胞分化后各基因mRNA表达的相对灰度值为Leptin0.82±0.09、Adiponetic0.79±0.05、PPARγ0.62±0.05、TSHR0.86±0.08、Pref-10.70±0.04,其中前4个基因表达明显较分化前增强(P<0.05),Pref-1则未见统计学差异。结论TAO患者眼眶前脂肪细胞在一定条件下可向成熟脂肪细胞转化,并伴随着相关基因的改变,在TAO的发生、发展中可能起着重要作用。Objective To discuss the adipose-related genes changes before and after orbital adipocyte differentiation in thyroid-associated ophthalmopathy (TAO). Methods Preadipocytcs from 14 TAO patients' orbits were cultured and differentiated into mature adipocyte. Using RT-PCR detection,the different gene expression were compared before and after differentiation, such as transcription factors PPARγ,Pref-1, adipocytokines Leptin,Adiponectin and TSHR. Results The relative gradations of mRNA expression after preadipocyte differentiation were Leptin 0.82 ± 0.09, Adiponetic 0.79 ± 0.05, PPARγ0.62 ± 0.05, TSHR 0.86 ± 0.08 ,Pref-1 0.70 ± 0.04. Stronger PPARγ, Ieptin,Adiponectin, TSHR mRNA expression were showed after adipose differentiation ( P 〈 0.05 ). There were no significant change in Pref-1 mRNA expression. Conclusion The preadipocytes in the orbit of TAO can be differenttated into mature lipid-filled adiocytes in appropriate medium, followed the related genes changes, which may make important effects on the development of TAO.
