魔芋甘露聚糖对运动应激大鼠肠粘膜屏障的保护作用

摘　　要：目的:探讨魔芋甘露聚糖(KOS)对运动应激大鼠肠粘膜损伤的保护作用。方法:32只SD大鼠随机分为安静对照组(n=7)、安静+KOS组(n=7)、力竭运动组(n=9)和力竭+KOS组(n=9),两运动组进行为期8周、每周6次、每次150min的游泳训练,两KOS组每天以0.90g/kg的剂量灌胃补充魔芋甘露聚糖,8周后测定各组大鼠肠粘膜屏障、肠粘膜形态学等参数,并观察肠组织病理变化。结果:8周大负荷训练后,大鼠肠粘膜的血浆内毒素、肠粘膜通透性和细菌移位率升高,肠液IgA浆细胞个数和SIgA含量下降,腺隐窝深度变浅;连续8周补充KOS的运动大鼠血浆内毒素、肠粘膜通透性、细菌移位率显著低于力竭运动组,肠粘膜厚度、腺隐窝深度、绒毛宽度、肠液IgA浆细胞个数和SIgA含量则显著高于力竭运动组。结果提示:运动应激导致的机体胃肠综合征可能与肠粘膜屏障的损伤有关,运动应激后补充KOS可以保护肠粘膜。Objective To explore the protective effects of konjacmannan （KOS） on the injured in- testinal mucous membrane caused by exercise stress. Methods Thirty two SD rats are randomly divided into the sedentary control group （C） （n=7）, the sedentary KOS group （C＋KOS） （n=7）, the exhaustive exercise control group （EE） （n=9） and the exhaustive exercise＋KOS （EE＋KOS） group （n=9）. In the exercise groups, rats were forced to swim 150min/day, 6 days/week for 8 weeks. The parameters of the intestinal mucous membrane barriers and membrane morphology were monitored, and the pathological changes in the small intestines were observed. Results After heavy training for 8 weeks, the endotoxin in blood, mucosa permeability, ratio of bit shifting of bacteria increased, while the numbers of IgA cells in intestine fluid, SIgA contents and the depth of gland crypt decreased. After continuously supplementation of KOS, the endotoxin in blood, mucosa permeability, and ratio of bit shifting of bacteria in group EE＋KOS were significantly lower than that in group EE, while the thickness of intestinal mucosa, depth of gland crypt, width of villus, the numbers of IgA cells in intestine fluid, and SIgA contents significantly higher in group EE＋KOS than that in group EE. Conclusion KOS revealed a protective effect on the injured intestines mucous membrane in rats caused by exercise stress.
