摘 要:目的应用基因重组获得人小肠三叶因子(hITF).并研究外源性hITF保护肠黏膜的作用及其可能机制。方法构建毕赤酵母重组表达质粒pPIC9/hITF。经发酵上清过柱纯化,应用质谱技术鉴定所得目的蛋白;用大鼠小肠隐窝上皮细胞株(IEC-6)观察不同浓度重组hITF对肠上皮细胞形态、增殖和迁移功能的影响。结果获得基因重组hITF,质谱分析证实为所需目的蛋白。体外实验结果表明,在10^(-9)、10^(-7)、10^(-5)mol/L浓度,重组hITF能促进肠上皮细胞迁移,但对肠上皮细胞的光镜下形态和增殖功能无明显影响。结论重组hITF能促进肠上皮细胞迁移,可能是其对肠黏膜保护的主要作用。Ohjectiv To construct human intestinal trefoil factor(hITF) by recombinant technique and to study the effect and potential mechanism of hITF in protection of intestinal epithelial cells. Method The recombinant hITF plasmid was constructed with a yeast-based expression vector pPIC9 in Pichia pastoris, and then purified from the yeast fermentation broth by anion- and cation-exchange chromatography. The parified protein was analysed by electrospray mass spectrometry. The effects of recombinant hITF (10^-9-10^-5mol/L) on enterocyte (IEC-6) morphology, proliferation and migration were examined. Results The recombinant hITF was successfully constructed, and verified by Moldi-TOF- TOF analysis. The recombinant hITF promoted cell migration in a concentration range of 10 9 to 10-5 mol/L, but had no effet on cell morphology and proliferation. Conclusion The recombinant hITF protects intestinal mucosa by promoting cell migration.
