摘　　要：探讨人转化生长因子β１对肿瘤细胞的影响。方法通过构建人转化生长因子β１（ｈＴＧＦβ１）基因表达载体，利用基因转染技术，将基因导入ＨＬ－６０细胞中，经Ｇ４１８选择获得可稳定表达人转化生长因子β１的ＨＬ－６０细胞，对该细胞进行了一系列恶性表型变化的研究。结果表达人转化生长因子β１ｍＲＮＡ的ＨＬ－６０细胞其生长速度下降，在软琼脂中集落形成能力降低，在裸鼠体内的成瘤性下降，与对照组比较，转染组裸鼠没有发生肝转移。结论ｈＴＧＦβ１对肿瘤细胞ＨＬ－６０的恶性表型具有明显的抑制作用。Objective To study the effect of transforming growth factor β 1 on tumor cells.Method HL 60 cells were transfected with transforming growth factor β 1 expression vector by electroporation.Stable cell clones resistant to G418were obtained after2 3week growth with the drug containing medium.A series of malignant phenotype were compared between transfected and untransfected HL 60cells.Results The growth speed of HL 60cells which overexpressed transforming growth factor β 1 mRNA decreased significantly.Clonigenecity in soft agarase and tumor formation in athymic mice were lower thanthose untransfected HL 60 cells.Conclusion Suppression effect of transforming growth factor β 1 on malignant phenotype in HL 60cells were demonstrated.
