垂体肿瘤转化基因和碱性成纤维细胞生长因子在胶质瘤中的表达及意义

摘　　要：目的探讨垂体瘤转化基因(PTTG)和碱性成纤维细胞生长因子(bFGF)在胶质瘤中表达及二者相互关系。方法40例星形细胞胶质瘤患者手术标本,病理学分级为Ⅰ级8例、Ⅱ级10例、Ⅲ级13例、Ⅳ级9例。逆转录聚合酶链式反应(RT-PCR)检测PTTG mRNA及bFGF mRNA的表达,免疫组化检测二者蛋白表达。结果PTTG mRNA、bFGF mRNA及蛋白表达均随着肿瘤病理级别的增高而增加(P<0.05或P<0.01);PTTG蛋白表达与bFGF蛋白表达呈显著正相关(r=0.523,P<0.05)。结论PTTG和bFGF与胶质瘤的生物学行为有密切关系,二者共同促进胶质瘤的发生发展。Objective To investigate the expression of pituitary tumor transforming gene （PTTG） and basic fibroblast growth factor （bFGF） in gliomas and detect the relationship between them. Methods Reverse transcription polymerase chain reaction （ RT - PCR） was used to detected the expression of PTTG mRNA and bFGF mRNA in the specimens from 40 patients with glioma, invcluding 8 cases of grade Ⅰ , 10 grade Ⅱ , 13 grade Ⅲ and 9 grade Ⅳ. The proteins expressed by PTTG and bFGF were detected by immunohistochemistry using streptavidin - peroxidase （SP） mothed. Results The expressions of PTTG mRNA and bFGF mRNA were both correlated to the pathological grade of glioma （ P 〈 0.05 or P 〈 0.01 ）. The expression of PTTG protein was positively correlated with that of bFGF protein （ r = 0.523, P 〈 0.05）. Conclusion The abnormal expression of PTTG mRNA and bFGF mRNA play important roles in the biobehaviors of glioma.
