摘 要:以鸡GARS-AIRS-GART基因为候选基因,根据5′侧翼序列设计引物,用PCR-SSCP方法对隐性白羽鸡、丝羽乌骨鸡、萧山鸡、白耳鸡及藏鸡进行单核苷酸多态性分析,并检测到多态性,表现为3种基因型,对2种纯合子进行直接测序,结果发现-179位点发生了突变,由C突变为T。对3种基因型的肌肉肌苷酸(IMP)含量采用最小二乘法分析,结果显示TT型(突变型)个体的肌肉IMP含量极显著高于CT型、CC型个体,CT型个体稍高于CC型,但差异不显著,初步推测该位点可能与肌肉IMP含量有关,可作为肌肉IMP含量的辅助选择标记。This study was designed to investigate the single nucleotide polymorphism (SNP) of GARS-AIRS-GART gene in various breeds, including Recessive White chickens, Silkies chickens, Xiaoshan chickens, Baier chickens and r Tibetan chickens. The primers for 5′ flanking in GARS-AIRS-GART gene were designed from the database of chicken genomic sequence and the SNPs were detected by PCR-SSCP and DNA sequencing. Three genotypes were found among individuals within all breeds and one SNP (C→T at -179) was found. However, the amino acid was not changed because it was a silence mutation. The least square analysis showed that TT genotype birds had significant higher (P〈0.01) IMP content than TC and CC genotype birds, TC genotype birds also had a little higher IMP content than CC genotype birds, but the difference was not significant. We concluded that there may be a relationship between this SNP site and IMP content in chickens.
