摘  要:目的研究水飞蓟宾(Silibinin,SB)在体外对人膀胱癌细胞株5637增殖的抑制作用及其相关机制。方法采用MTT法观察不同浓度SB对人膀胱癌细胞株5637增殖的抑制作用;倒置显微镜下观察细胞形态变化;RT-PCR检测凋亡抑制因子Survivin的mRNA表达变化。结果MTT检测发现:随着SB浓度增加和作用时间延长,对人膀胱癌细胞株5637增殖的抑制作用增强,且各组间比较均有显著性差异(P<0.01),并呈时间-剂量依赖关系;倒置显微镜发现应用SB干预后,5637细胞体积缩小,变形,胞浆浓缩,核固缩深染;人膀胱癌细胞株5637中Survivin的mRNA表达随着SB浓度增加和时间延长相应下降。结论SB可抑制人膀胱癌细胞株5637的增殖;并下调凋亡抑制因子Survivin的mRNA表达。Objective To determine the effects of silibinin on inhibiting the proliferation of 5637 cell in vitro and to investigate the related mechanism. Methods The cell growth activity was evaluated by MTT assay. Morphological changes were studied through the inverted phase contrast microscope. The expression of survivin (inhibitor of apoptosis protein) mRNA was determined by reverse transcription-polymerase chain reaction (RT- PCR). Results Silibinin treatment of 5637 cells resulted in a moderate to strong growth inhibition in both a doseand a time-dependent manner. There were significant differences between these groups (P 〈 0. 01 ). These morphological changes of 5637 which were treated with silibinin: cells shrank and turned round, condensed cytoplasma and pycnosised nucleus were observed through the inverted phase contrast microscope. There was also significant difference in the positive expression of survivin mRNA between the 5637 cell treated by differentiated silibinin. Oonclusion Silibinin could inhibit the growth of human bladder cancer 5637 cells and down-regulate the expression of survivin.
