摘 要:目的探讨脑梗死对成年小鼠室管膜下区(SVZ)神经发生的影响。方法将48只小鼠随机分为假手术组和脑梗死组(各24只),建立局灶性大脑中动脉闭塞(MCAO)模型。采用5-溴脱氧尿核苷(BrdU)标记神经干细胞,用免疫组化法观察脑梗死后7、14、21及28d时,损伤侧SVZ区、纹状体、皮质BrdU阳性细胞数及BrdU分别与微管相关蛋白Doublecortin(DCX)、胶质纤维酸性蛋白(GFAP)、神经元核心抗原(NeuN)双染细胞数的变化。结果MCAO术后7d,脑梗死组损伤侧SVZ区、纹状体、皮质BrdU阳性细胞明显增多,假手术组与脑梗死组分别为45.0±5.6和245.0±26.1、0和201.9±21.3、0和159.3±16.4,14d后开始下降(P<0.05)。脑梗死组MCAO术后7和14d,可在小鼠损伤侧SVZ区、纹状体观察到BrdU和DCX阳性细胞;术后28d,损伤侧纹状体可见BrdU和NeuN阳性细胞,数量均显著多于假手术组(P<0.05),BrdU和GFAP阳性细胞数差异无统计学意义。结论脑梗死可促进成年小鼠损伤侧SVZ区、纹状体及皮质的BrdU阳性细胞数增多,并诱导SVZ区干细胞向纹状体迁移且分化为神经细胞,从而促进SVZ区的神经发生。Objective To investigate the influence of cerebral infarction on neurogenesis in the subventricular zone (SYZ) of adult mice. Methods Forty-eight mice were randomly assigned to a cerebral infarction group ( n = 24) and a sham-operation group ( n = 24). A model of focal middle cerebral artery occlusion (MCAO) was established. Neural stem cells were labeled by 5-bromo- 2′-deoxyuridine 5′-monophosphate (BrdU). The SVZ, striatum and number of cortical BrdU-positive cells on the affected sides at day 7, 14, 21 and 28 after MCAO, as well as BrdU and the changes of the number of amphophilic cells in doublecortin (DCX), glial fibrillary acidic protein (GFAP) and neural nuclear protein (NeuN) were detected by immunohistochemistry. Results Compared with the sham operation group, the SVZ, striatum and number of cortical BrdU- positive cells on the affected sides increased significantly 7 days after MCAO in the cerebral infarction group, and they were 45.0 ±5.6 vs 245.0 ±26. 1,0 vs 201.9±21.3 and 0 vs 159. 3 ±16.4, respectively. They began to decrease 14 days later ( P 〈 0. 05 ). BrdU and DCX positive cells were observed in SVZ and striatum on the affected sides 7 and 14 days after MCAO procedure in the cerebral infarction group. BrdU and NeuN positive cells were observed in striatum on the affected sides 28 days after MCAO procedure; and the numbers were significantly higher than the control group. Conclusion Cerebral infarction may promote the increase of the SVZ, striatum and cortical BrdU-positive cells on the affected sides in adult mice, and induce stem cells of SVZ to migrate to striatum and differentiate into neurons, thus enhance neurogenesis in SVZ.
