摘  要:目的探讨小鼠Lewis肺癌(Lewis lung carcinoma,LLC)细胞株中Sca-1+细胞的成瘤特性。方法以Sca-1作为磁珠分选细胞的表面标志,从LLC细胞中分离Sca-1+细胞,流式细胞仪鉴定分离的纯度。将分选的Sca-1+细胞、Sca-1-细胞及未分选细胞分别以1×106、5×105、1×105、5×104、2×104密度接种于C57BL/6小鼠腋窝下,观察成瘤时间,4周后处死小鼠,统计成瘤率、平均瘤重及体积。结果分选前Sca-1阳性细胞百分比为(8.06±1.03)%,分选后Sca-1阳性细胞百分比为(93.92±1.07)%。Sca-1+细胞组在2×104个细胞时可以成瘤,而Sca-1-细胞组最少需要5×105个细胞。相同接种量(1×106),4周后处死小鼠,Sca-1+细胞组的瘤重及体积均大于Sca-1-细胞组(P<0.01)。结论Sca-1+细胞的成瘤性较Sca-1-细胞强。Objective To explored the tumorigenicity of sca-1 positive cells from Lewis lung carcinoma Cell Line in C57BL/6 Mice. Methods The sea-1 positive cells were sorted by magnetic activated cell sorting (MACS) from Lewis lung carcinoma Cell Line, the purity was analyzed by the method of fluorescence-actived cell sorting (FACS). Unseparated, Sca-1^+ and Sca-1^- populations were inoculated into the axillary fossa of C57BL/6 mice at the injection dose 1 × 10^6 , 5 × 10^5 , 1 × 10^,5 × 10^4 ,2 × 10^1 respectively. The mice were monitored for palpable tumor formation and were sacrificed after 4 weeks to assess tumor formation rate, volume and weight. Results FACS analysis shown the Sca-1 positive fraction was (8.06 ± 1.03) % in Lewis lung carcinoma Cell Linc. After isolation, the sca-1 positive fraction was raised to (93.92±1.07)%. Sca-1^+ cells generated a tumor with 2 × 10^4cells compared with at least 5 × 10^5 needed for Sca-1^- cells, at the same injection dose ( 1 × 10^6cells) , the tumor generated by the Sca-1^+ cells was larger than that of the Sca-1^- cells. Conclusion Sca-1^+ cells have higher tumorigenicity than Sca-1^- cells.
