摘 要:目的探讨壳聚糖诱导子对地钱细胞生长和酚类次生代谢的影响。方法对壳聚糖-地钱细胞诱导体系的细胞生物量、叶绿素、活性氧(H2O2和O2-)、细胞膜通透性和膜脂过氧化产物(MDA)以及酚类次生代谢物苯丙氨酸解氨酶(PAL)和多酚等生化指标进行测定。结果细胞生长下降(P<0.05),叶绿素含量明显低于对照组(P<0.01),胞外pH趋向碱性化(P<0.01),活性氧含量急剧增加和膜脂过氧化增强(P<0.01),次生代谢PAL活性和多酚含量均显著增加(P<0.01)。结论壳聚糖通过降低地钱细胞还原能力而增强其氧化能力,从而有利于酚类次生代谢的生物合成。Objective To study the role of the chitosan elicitor on cell growth and phenolic compound contents of Marchantia polymorpha L. Methods: Cell biomass, chlorophyll, reactive oxygen species (H2 02 and O^-2 ), cell membrane permeability and membrane lipid peroxidation production (MDA) and phenolic secondary metabolism (phenylalanine ammonialyase and phenolic compounds) were investigated in suspension cultures of Marchantia polymorpha induced by the chitosan elicitor. Results Cell biomass decreased ( P 〈 0.05 ), and chlorophyll content also decreased ( P 〈 0.01 ), while pH of the medium tended to be alkali( P 〈 0.01 ). Reactive oxygen species greatly increased ( P 〈 0.01 ) and membrane lipid per-oxidation markedly intensified ( P 〈 0.01 ). Meanwhile, the PAL activity and phenolic compounds of secondary metabolism had a significant increase (P 〈 0.01). Conclusion The ability to de-xidation decreases and an oxidation increase of Marchantia polymorpha cells induced by chitosan may favor phenolic compounds biosynthesis.
