摘 要:目的:以往研究证实,针刺丰隆穴可有效的调节血脂,为了进一步研究其对高脂血症后肝脏脂肪变性的作用机制,实验观察了电针丰隆穴对高脂血症大鼠脂蛋白脂酶、肝脂酶活性及肝细胞脂肪变性的影响。方法:实验于2005-09/12在北京中医药大学针灸学院综合实验中心完成。①实验干预:选用健康雄性SD大鼠30只,鼠龄4个月。随机分为3组:空白组、模型组、丰隆穴组,每组10只。空白组每日给予基础饲料喂养及普通饮水;其余两组均给予高脂饲料喂养及普通饮水2周以制备高脂血症模型。造模2周后开始电针干预。丰隆穴组:取双侧丰隆穴,直刺10mm,接电针仪,频率50Hz,强度1mA,留针20min,每周治疗2次,治疗10次。空白组、模型组于丰隆穴组治疗时同样抓取,但不针刺。②实验评估:治疗结束后,所有大鼠腹腔静脉取血,采用分光比色方法测定血清脂蛋白脂酶、肝脂酶活性。摘取肝脏进行油红"O"染色,并光镜下观察肝脏脂肪细胞组织形态学表现。结果:大鼠30只均进入结果分析。①血清脂蛋白脂酶、肝脂酶活性:模型组与空白组比较,差异无显著性意义(P>0.05);丰隆穴组高于模型组,差异有十分显著性意义(P<0.01)。②肝脏脂肪细胞组织形态学表现:模型组肝细胞明显增大,且排列松散紊乱;肝组织的汇管区、中央静脉区等细胞内可见数量众多、颜色深染、体积较大的红染脂滴,而且肝小叶结构被破坏。丰隆穴组比模型组的肝脂肪细胞小、数量少、染色浅。结论:电针丰隆穴可提高高脂血症大鼠血清脂蛋白脂酶、肝脂酶活性,减轻肝细胞内脂肪的堆积,对肝脏脂肪变性有改善作用。AIM: It has been proved in the former studies that acupuncture at Fenglong (ST.40) could effectively regulate blood fat. In this study, the effect and mechanism of the electro-acupuncture on Fenglong on lipoprotein lipase (LPL), hepatic lipase (HL), and hepatic steatosis in rats with hyperlipemia were investigated. METHODS: The experiment was conducted in the Experimental Center, College of Acupuncture and Moxibustion, Beijing University of Chinese Medicine from September to December 2005. (1)Thirty healthy male SD rats of 4 months old were selected and randomly divided into 3 groups with 10 rats in each group: blank control group, model group, and Fenglong group. Rats in the blank control group were fed with basic diet everyday, while the rest groups were fed with high fat diet everyday for 2 weeks to establish the models of hyperlipemia. Two weeks after modeling, the rats in the Fenglong group were treated with electro-acupuncture at bilateral Fenglong for 10 mm, 50 Hz, 1 mA for 20 minutes, twice every week for 10 times. (2)After treatment, the blood samples were drawn from abdominal vein to test LPL and HL by spectrocolorimetry. At the same time, liver was harvested for oil red O staining test to observe the histomorphology of fatty cells under light microscope. RESULTS: Thirty rats were involved in the result analysis. (1)There was no significant difference in LPL and HL activity between model group and blank control group (P 〉 0.05), while the LPL and HL in Fenglong group was remarkably higher than that in model group (P〈 0.01). (2)In the model group, the fatty cells of liver were markedly increased with confused arrangement, and many deeply stained large red lipid droplets were found in the portal area and central veins; moreover, hepatic Iobules were damaged. In Fenglong group, the fatty cells were less, smaller and light-colored than in model group. CONCLUSION: Electro-acupuncture at Fenglong could increase the activity of LPL and HL, relieve fatty accumul
