摘 要:目的探讨叶绿素显示液对胃淋巴结显色的应用价值。方法应用光、电镜技术和生化分析技术研究注射叶绿素显示液后,狗胃淋巴结染色情况及其在血液中浓度变化情况。结果注射叶绿素显示液后,平均每只狗观察到淋巴结个数明显高于注射叶绿素之前发现淋巴结个数(P<0.01);光镜下叶绿素进入淋巴结的被膜下窦和小梁周窦;随着时间延长,淋巴结髓质内亦可见叶绿素颗粒,但未见到细胞结构出现异常情况;电镜下淋巴结皮、髓质内均有叶绿素颗粒,以单核细胞和巨噬细胞内多见,但未发现淋巴结细胞超微结构发生改变;血液中叶绿素浓度于注射后4h达高峰,之后逐渐减少,24h后叶绿素在血液内基本消失。结论叶绿素显示液是一种理想的淋巴结显色物质,该方法可明显提高胃淋巴结的检出率及避免损伤周围组织。Objective To investigate the application value of chlorophyll disclosing solution in coloration of dog gastric lymphonodes. Methods The coloration conditions of dog gastric lymphonodes were investigated under light and electron microscopy. The concentration of chlorophyll in the blood was measured by biochemistry analytical technique. Results After chlorophyll was injected into the gastric wall of dogs, The number of lymph nodes increased obviously compared with the mum ber bafore injection (P〈0.01); Chlorophyll grains were found in subcapsular sinuses and trabeculum sinuses and were also observed in medullary sinus and efferent lymphatics with time passed. No abnormal cellular structure was observed in the experiment; Chlorophyll grains were observed in cortex and medulla of lymph nodes under the electron microscope, and most of them located in lymphomonoeyte and macrophagocyte. No change on ultrastructure of lymph nodes was found in study group. The concentration of chlorophyll in the blood was maximum at 4th hour after injection, and decreased gradually with time passed and eliminated at 24th hours. Conclusion The chlorophyll disclosing solution was a useful dye for staining lymph nodes. The method may improve the detection rate of stomach lymph nodes obviously and avoid injuring adjacent tissues.
