胃癌和相应癌旁组织中PTEN基因表达的临床研究

摘　　要：目的研究胃癌组织中抑癌基因第10号染色体缺失性磷酸酶张力蛋白基因(phosphatase and tension homologydeleted on chromosome ten,PTEN)的表达情况及探讨PTEN基因与胃癌临床病理因素的关系。方法应用半定量逆转录聚合酶链反应(semi-quantitative RT-PCR)方法检测45例胃癌组织及相应癌旁组织中PTEN基因1-8、5-85、-9 mRNA的表达情况。结果45例胃癌组织中PTEN mRNA表达水平为:外显子1-8为0.71±0.21;外显子5-8为0.90±0.11;外显子5-9为0.83±0.15;45例相应癌旁组织中PTEN mRNA表达水平为:外显子1-8为1.29±0.18;外显子5-8为1.51±0.27;外显子5-9为1.38±0.22;两者相比差别显著(P<0.05);而45例相应癌旁组织中PTEN mRNA全部阳性表达,与相应胃癌组织相比差别显著(P<0.05)。胃癌组织中PTEN mRNA表达水平无性别、年龄特异性(P>0.05),但随临床分期的增高和组织学分化的降低,其表达水平明显降低(P<0.05)。结论PTEN基因在胃癌中表达降低和缺失与胃癌的发生密切相关,并与临床分期与病理分级的进展密切相关,提示PTEN mRNA的表达有望作为胃癌诊断的一项客观指标。Objective To investigate the expression of PTEN （phosphatase and tension homology deleted on chromosome ten） mRNA in gastric carcinomas and their adjacent tissues as well as to appreciate its clinical significance. Methods The relative mean expression of PTEN mRNA in these samples was detected by semi-quantitative RT-PCR.Results Of the 45 gastric carcinomas, the relative mean expression of PTEN mRNA to β-actin was exon 1-8 0.07 ±0.21,5-8 0.09± 0.11,exon 5-9 0.83 ± 0.15.of the 45 gastric carcinomas adjacent tissues,the relative mean expression was exon 1 - 8 1.29 ± 0.18,exon 5 - 8 1.51 ± 0.27,exon 5 - 9 1.38 ± 0.27. However, the positive of PTEN mRNA was found in all of the 45 gastric carcinomas adjacent tissues. The expression of PTEN mRNA did not show correlation with such faction as sex,age,but had much to do with the clinical staging and histological grading. Conclusion The expression level of PTEN mRNA in gastric carcinomas was sharply reduced compared with their adjacent tissues, with suggests that PTEN PTENA inactivation plays an important role in the genesis and development of gastric carcinomas. The expression of PTEN mRNA was expected to be a diagnostic criteria of gastric carcinomas.
