恶性脑肿瘤浸润淋巴细胞的临床应用研究

摘　　要：用冷酶法对７例脑恶性肿瘤进行肿瘤浸润淋巴细胞分离、培养并用于临床。７例肿瘤的ＴＩＬ扩增倍数均大于刚分离时的５００倍，其中２例大于１０００倍，另２例分别大于２０００及３０００倍；ＴＩＬ的表型检测ＣＤ３、ＣＤ４、ＣＤ８、ＨＬＡ－ＤＲ分别为９７％，３５％，５４％，９７％；电镜观察到ＴＩＬ先靠近肿瘤细胞，伸出触手，接触肿瘤细胞膜上，随后肿瘤细胞破坏、死亡的过程。临床应用ＴＩＬ后随访至今９ｍｏ，７例肿瘤患者中，１例转移性腺瘤因全身衰竭于手术后５ｍｏ死亡，另１例术后６ｍｏ复发，其他患者无复发。We isolated and cultured tumor infiltrating lymphocytes from seven brain cancer patients by cold digestion method and investigated their clinical usage Growth of TILs all increased 500 fold,tow of them increased more than 1 000 times,and other two of them 3 000 times,respectively The phenotypies of TIL CD 3,CD 4,CD 8,HLA DR were 97%,35%,54%,97% We observed by microscope that TILs went close to tumor cells,touched the membrane by stretching their tentacle,and then destroyed tumor cells We had followed up the seven patients after TILs infusion for nine months One patient died of the whole body exhanstion at the fifth month,one patient had a relaps at the sixth month,and the others had no relaps
