摘 要:目的:检测BRCA1、Bad在乳腺导管普通性增生、不典型增生、乳腺癌组织中的表达,探讨BRCA1、Bad在乳腺癌发生、发展中的作用及其意义。方法:应用免疫组化SP法检测20例乳腺导管上皮普通性增生、28例乳腺导管上皮不典型增生、25例乳腺癌中BRCA1、Bad表达情况。结果:(1)BRCA1在乳腺导管普通性增生、不典型增生、乳腺癌组织中的阳性表达率分别为5%(1/20)、39.3%(11/28)、76.0%(19/25),三者比较,差异均具有显著性(P<0.01)。(2)Bad在三者中的阳性表达率为分别15%(3/20)、35.7%(10/28)、72.0%(18/25),Bad蛋白在普通性增生组织和不典型增生组织的阳性表达率差异无统计学意义(P>0.05)。在普通性增生组织和乳腺癌组织、不典型增生组织和乳腺癌组织之间的阳性表达率差异有统计学意义(P<0.01)。(3)BRCA1、Bad在乳腺病变中的表达不具有相关性。结论:提示BRCA1、Bad表达异常与乳腺癌的发生、发展有关,可能是乳腺癌形成和发展的重要机制。Objective: To investigate the expression of BRCA1 ,Bad in breast benign and maligant lesions and the correlation to cancer development. Method: Immunohistochemical staining method of SP were used to detect the BRCA1, Bad gene expressions in 20 usual ductal hyperplasia, 28 atypical ductal hyperplasia and 25 breast cancer tissue. Result: (1) immunohistochemical staining method of SP results revealed that when compared with the positive rates of usual ductal hyperplasia, atypical ductal hyperplasia and breast cancer were5%(1/20), 39.3%(11/28), 76.0%(19/25) respectively inBRCA1. The positive rates of BRCA1 in these three were significant (P 〈0.01). (2) The Bad positive rates were 15.0%(3/20), 35.7%(10/28), 72.0%(18/25) respectively. The positive rates of Bad in usual ductal hyperplasia and atypical ductal hy- perplasia were insignificant (P〉0.05). The positive rates of Bad in atypical ductal hyperplasia and breast cancer, in usual ductal hyperplasia and breast cancer were significant (P 〈0. 01). (3) Expression of BRCA1, Bad were no relation in the breast disease. Conclusion : The abnormal expressions of BRCA1, Bad gene may be related to the development of breast cancinoma.
