摘　　要：目的探讨选择性环氧合酶-2(COX-2)抑制剂Celebrex对人胆囊癌GBC-SD细胞系血管内皮生长因子(VEGF)表达的影响。方法采用RT-PCR、免疫细胞化学技术以及Western blot联合检测Celebrex对GBC-SD细胞系VEGF mRNA和蛋白表达的影响,并观察量效和时效关系。结果随着药物浓度的增加(0、40、80、120、160μmol/L)和作用时间的延长(0、12、24、48、72h),VEGF mRNA和蛋白表达均呈现出了相应的下降趋势。结论Celebrex可以有效地抑制人胆囊癌GBC-SD细胞系VEGF的表达。Objective To investigate the effects of cyclooxygenase-2 inhibitor celebrex on the expression of vascular endothelial growth factor（VEGF） in gallbladder carcinoma GBC-SD cell line.Methods The concentration-effect relationship and time-effect relationship between celebrex and the expression of VEGF in gallbladder carcinoma GBC-SD cell line were observed by using immuocytochemistry,reverse transcription polymerase chain reaction（RT-PCR） and Western blot.Results With the increase of celebrex concentrations（0,40,80,120 and 160 μmol/L） and prolongation of time（0,12,24,48,72 h）,the expression of VEGF mRNA and protein was decreased accordingly.Conclusion Celebrex may play an important role in inhibiting the expression of VEGF in gallbladder carcinoma GBC-SD cell line.
