摘  要:目的研究诱导型一氧化氮合酶(iNOS)在兔碱烧伤角膜的表达与角膜新生血管形成的关系。方法将33只家兔分为正常组(3只,6眼)、烧伤组(30只,60眼),以2mol/LNaOH制备双眼角膜重度碱烧伤模型,用裂隙灯图像分析系统记录角膜新生血管(CoNV)生长情况。碱烧伤组分批(n=5)于伤后第1、4、7、14、21、28天处死家兔,分别取角膜组织行病理学检查和iNOS的免疫组织化学检测。结果iNOS在正常兔角膜上皮层及基质层仅有少量表达,在烧伤组表达明显增强(P<0.05),表达范围主要在基质层炎性细胞、角膜上皮细胞及新生血管内皮细胞,表达峰值在术后第7天,随后下降,在第21天时又有所回升。结论碱烧伤后角膜组织中iNOS的表达明显增加,其变化趋势与碱烧伤后角膜新生血管的生长曲线相一致且高峰期提前2~3d,提示iNOS水平与无菌性炎症性新生血管形成有明显的相关性。Objective To study the relationship between the expression of inducible nitric oxide synthase and corneal neovascularization(CoNV) in rabbits with corneal alkali burns.Methods Thirty-three rabbits were divided into normal control group(n=3,6 eyes) and burns group(n=30,60 eyes).The severe corneal alkali burn model in rabbits was established with 2 mol/L NaOH.Digital slit lamp image system was used to observe the newly formed vessels.The rabbits in burns group were killed at 1st,4th,7th,14th,21st,28th day respectively after burns(n=5,10 eyes in each time).Corneal samples were harvested for histopathological examination and immunochemical analyses of iNOS.Results In the normal rabbits corneas slightly expressed iNOS,but in the burns group,the expression of iNOS was significantly increased(P〈0.05).Conclusion iNOS is expressed remarkably in this model of sterile corneal inflammation after alkali burn.Moreover,the change tendency is in coincident with the CoNV,and the peak is earlier 2-3 days than the CoNV growth peak.These results indicate that the levels of iNOS are markedly related with sterile inflammatory CoNV.
