摘  要:目的:探讨雷公藤内酯醇(triptolide,Tri)抑制体外培养的牛晶状体上皮细胞(bovine lens epithelialcell,BLECs)增殖及其机制。方法:取第4代BLECs,用MTT比色法检测增殖抑制率来测定Tri 20μg/L对细胞增殖的影响,用放射免疫法测定BLECs内cAMP浓度。结果:Tri 20μg/L,时间6,12,24,48,72 h作用在处于增殖状态BLECs,出现明显的增殖抑制作用,其增殖抑制率升高呈现明显的时间依赖性。而增殖抑制组细胞内cAMP含量明显增加。结论:Tri对晶状体上皮细胞增殖有明显的抑制作用,而该作用可能通过影响细胞内cAMP等第二信使物质浓度来完成。Objective: To investigate the inhibitory effects on proliferation of cultured bovine-lens epithelial cells(BLECs) in vitro and the mechanism by Triptolide.Methods: The fourth generation of BLECs was used for the experimental research.The inhibitory rates of BLECs were measured by MTT(colorimetric assay),the intracellular cAMP was measured by Radioimmunoassay.Results: The distinct inhibitory effects on the proliferation of BLECs in vitro were clearly observed and measured by Triptolide with 20 μg/L concentrations at the different time lengths.The inhibitory rates increased in the time-dependent way.The concentration levels of intracellular cAMP were evidently increased at the same time of producing inhibitory effects. Conclusion: Triptolide could effectively inhibit the proliferation of BLECs in vitro.The triptolide inhibition on the proliferation may accomplished by means of changing the concentration of the intracellular cAMP.
