摘 要:目的探讨异丙酚联合电休克治疗对抑郁大鼠行为学和学习记忆功能的影响。方法成年健康雄性SD大鼠50只,随机分为5组(n=10):对照组(C组)、抑郁模型组(D组)、电休克组(E组)、异丙酚联合电休克组(M组)和异丙酚组(P组)。C组正常喂养,其余组建立抑郁模型。造模后D组正常喂养,E组和M组分别给予单纯电休克和异丙酚联合电休克治疗各6次,P组腹腔注射异丙酚100 mg/kg。各组分别于造模前、造模后第2天、干预结束后第2天用旷场法进行行为学评分。干预结束后第2天Morris水迷宫法测试学习记忆功能。采用分光光度计检测海马组织一氧化氮合酶(NOS)活性和一氧化氮(NO)含量,RT-PCR法检测神经元型一氧化氮合酶mRNA(nNOS mRNA)表达。结果与C组比较,其余4组行为学评分均降低,学习记忆功能均减弱(P〈0.05)。与D组和E组比较,M组干预结束后第2天行为学评分升高,学习功能减弱,记忆功能增强,NOS活性、NO含量和nNOS mRNA表达降低(JP〈0.05)。与P组比较,M组干预结束后第2天行为学评分升高,学习功能减弱,记忆功能增强(P〈0.05)。结论异丙酚联合电休克治疗可明显改善抑郁大鼠的行为学和学习记忆功能,其对学习记忆功能的影响可能与异丙酚抑制电休克介导的海马组织nNOS mRNA的过度表达,降低NO生成过多所致的中枢神经毒性反应有关。 Objective To investigate the effect of propofol combined with electroconvulsive therapy (ECT)on behavior and learning ability and memory in mentally depressed rats and the possible mechanism. Methods Fifty adult male SD rats weighing 250-300 g were randomly divided into 5 groups(n=10 each);Ⅰcontrol group(C);Ⅱmentally depressed group(D);ⅢECT group(E);Ⅳpropofol+ECT(M)andⅤpropofol group(P).Mental depression model was established in groupⅡ-Ⅴby subjecting the animals to one of mild stress alternatively once a day for 21 consecutive days,including 2 h restraint,swimming in 4℃water for 5 min,24 h no food,24 h no water,clamping tail for 1 min,electric stimulation of plantar surface,tilling the cage, 45℃high temperature.In ECT group(groupⅢ)the animals received ECT 3 times a week for 2 weeks.In group M(groupⅣ)the animals received intraperitoneal propofol 100mg/kg+ECT 3 times a week×2 weeks and in group P(Ⅴ)the animals received propofol 100 mg/kg i.p.at the corresponding time points.The behavior of the animals was evaluated by open-field test and learning ability and memory by Morris water maze test before mental depression model was started(T0),2 days after the model was established(T1)and 2 days after treatment(T2). The animals were then anesthetized with intraperitoneal 10% chloral hydrate 3.5 nd/kg.The brains were immediately removed and the right and left hippocampi were isolated for determination of NOS activity and NO content by speetrophotometry.Results The behavior scores and learning ability and memory were significantly decreased after mental depression model was established in groupⅡ,Ⅲ,ⅣandⅤas compared with control group (Ⅰ).The behavior scores were significantly increased and memory was improved but the learning ability was decreased after treatment(at T2)in group M as compared with group D,E and P.The NOS activity,NO content and nNOS mRNA expression were significantly lower in group M and P than in group D and
