摘  要:目的探讨环氧化酶-2(COX-2)、诱导型一氧化氮合酶(iNOS)及血管内皮生长因子(VEGF)在星形胶质细胞瘤中的表达及意义。方法采用组织芯片及免疫组化技术检测12例正常脑组织、49例低级别星形胶质细胞瘤(Ⅰ-Ⅱ级)及50例高级别(Ⅲ-Ⅳ级)星形胶质细胞瘤中COX-2、iNOS及VEGF的表达。结果(1)在正常脑组织中三者均为阴性表达;随着星形胶质细胞瘤组织学分级的升高,COX-2、iNOS及VEGF的阳性表达率增加,分别为:COX-2:68%(32/47)、86%(36/42)、iNOS:58%(27/47)、82%(40/49);VEGF:44%(19/43)、70%(30/43)。(2)COX-2、iNOS及VEGF、在星形胶质细胞瘤中的表达有明显的相关性(P<0.05)。结论COX-2、iNOS及vEGF在星形胶质细胞瘤发生发展中起重要作用;COX-2、iNOS通过上调VEGF的表达来促进星形胶质细胞瘤血管形成。Objective To investigate the different expression of cyclooxygenase 2 (COX-2) inducible nitric oxide synthase (iNOS) and vascular endothelial growth factor(VEGF) in astrocytoma and their relationships in the stages of astrocytic tumorigenesis and growth. Methods Immunonhistochemistry and tissue microarray techniques were used to determinate the expression of the three biomarkers in 49 cases of low-grade astrocytomas and 50 cases of high-grade astrocytomas; meanwhile, 12 cases of normal brain tissues were used as control. Results ( 1 ) The expression of the three biomarkers was negative in the control group, but stepwise increase of the positive rate of expression of them was found from low-grade astrocytoma to high-grade astrocytoma, COX-2:68% ( 32/47 ) ,86 ( 36/42 ) ; iNOS: 58% ( 27/47 ) ,82% (40/ 49) ; VEGF: 44% ( 19/43 ) ,70% (30/43) ; respectively. (2) The expression of COX-2, iNOS and VEGF was positively correlated with each other in astrocytoma. Conclusions COX-2 iNOS and VEGF may play a great role in the stages of astrocytic tumorigenesis and growth; Overexpression of COX-2 and iNOS may contribute to VEGF-mediated angiogenesis of astrocytoma.
