摘 要:目的:观察153钐-氯屈膦酸二钠(153Sm-clodronate)在原位人骨肉瘤裸鼠动物模型中的疗效。方法:以人骨肉瘤细胞系HOS-8603接种SCID小鼠皮下,成瘤后经BALB/c裸鼠胫骨移植传代,再接种裸鼠胫骨建立原位人骨肉瘤裸鼠模型,测定153Sm-clodronate的生物分布,并通过外观、瘤体质量、钼靶X线片、组织学观察和生存期与153Sm-EDTMP、clodronate对照进行疗效观察。结果:注射后24 h生物分布示瘤骨分布远高于其他组织,放射性计数比瘤骨/血=4 627.1:1,瘤骨/正常骨=10.8:1,正常骨/肝脏=8.4:11。53Sm-clodronate治疗组较对照组溶骨反应轻,生存期显著延长,但是瘤体质量没有明显差异。结论:153Sm-clodronate浓聚于瘤骨,在原位人骨肉瘤裸鼠模型中的疗效较好,有进一步实验研究和临床应用的前景。Objective:To observe the therapeutic effect of 153 Sm-clodronate on nude mice orthotopically transplanted with human osteosarcoma. Methods: Severe combined immune deficiency (SCID) mice were subcutaneously inoculated with human osteosarcoma cell line HOS-8603. The formed tumors were then transplanted into BALB/c nude mice at the tibia for passage; the tumors were harvested for establishing orthotopical mice model of human osteosarcoma. Mice treated with 153 Sm-EDTMP and clodronate group served as controls. The distribution of 153Sm-clodronate was determined 24 h later. The appearance, tumor weights, X-ray examination, histological observation, and survival of mice were compared between the 3 groups. Results: It showed that the distribution of 153 Sm-clodronate in the lesion bone was much higher than that in other organs, with the radioactive ratio of lesion bone to blood being 4 627.1 : 1, lesion bone to normal bone being 10.8 : 1, and normal bone to liver being 8. 4 : 1. 15z Sm-clodronate treated mice had a slighter osteolytic reaction and a longer survival period, but the tumor mass was not significantly different compared with those of the control group. Condusion: 153 Sm-clodronate gathers at the lesion bone and it has good therapeutic effect on the mice model of human oseosarcoma, making it a promising agent for treatment of bone tumor.
