摘  要:目的检测胸腺五肽不同给药间隔对实验性自身免疫性重症肌无力小鼠(EAMG)T淋巴细胞亚群的影响。方法C57BL/6雌性小鼠随机分为正常对照组、模型对照组及3种不同给药间隔的治疗组,每组8只。治疗期内各治疗组给药总量相同,分别于模型成功后每1天(Ⅰ组)、每2天(Ⅱ组)或每4天(Ⅲ组)给药。结果荧光显微镜法检测CD4+/CD8+结果显示,正常对照组低于EAMG模型组或给药Ⅰ组(P<0.05),给药Ⅱ组低于EAMG模型组(P<0.05);给药Ⅲ组接近EAMG模型组(P>0.05)。结论提示胸腺五肽作为EAMG小鼠CD4+/CD8+的免疫调节剂,以每2天给药1次的效果较好。Purpose To probe the changes of CD4^+/CD8^+ in EAMG mice and in EAMG with Thymopentin (TP-5) therapy at different drug dispensing intervals. Methods Forty C57BL/6 mice were randomly divided into five groups-normal control group, EAMG group, EAMG with daily peritoneal injection of TP-5 (group Ⅰ ), EAMG with ip TP-5 every two days (group Ⅱ), and EAMG with ip TP-5 every four days (group Ⅲ ). The three treatment groups received the same amount of TP-5 through the experiment period. Their Tlymphocyte subgroups in peripheral blood were measured by fluorescence microscopic method. Results The value of CD4 ^+/CD8 ^+ in normal control group was significantly higher than that in EAMG group or group Ⅰ ( P 〈 0.05), yet similar to the value of group Ⅱ ( P 〉 0. 05). The value of CD4 ^+/CD8 ^+ in group Ⅱ was significantly different from that of EAMG group ( P 〈 0.05)while the value in group Ⅲ was not different from that of EAMG group( P 〉 0.05). Conclusion As a CD4 ^+/CD8^ + modulating agent used in MG therapy, TP-5 every two days showed a better efficacy than drug dispensed at every-day or every 4-day interval if weekly total dose remained the same for all. The results suggested that when immune-modulating agents are made into slow release for mulation, the considerations of drug dispense interval and dosage would be even more critical than that for some other types of drugs.
