摘  要:目的:观察PDT对SW480细胞和转染CD自杀基因的SW480结肠癌细胞周期和凋亡的影响。方法:采用脂质体基因转移技术将CD基因转染SW480结肠癌细胞后,进行PDT处理,用流式细胞仪分析ALA-PDT结合CD/5Fc自杀基因系统对结肠癌细胞周期和凋亡的影响。结果:ALA-PDT可使SW480细胞G1,细胞比率明显降低,S期细胞比率升高;ALA-PDT不会增加SW480细胞凋亡。PDT与5—Fc结合对SW480-CD细胞各周期比率的影响与对照组相似,表明对各期细胞影响度相似;两种方法结合可以明显诱导SW480细胞凋亡。结论:静止期SW480细胞对PDT比较敏感,增殖期细胞相对不敏感。PDT与5-Fc结合对静止期和增殖期癌细胞均有杀伤作用,且可明显诱导细胞凋亡。Objective To investigate the effects of cell cycle and apoptosis by photodynamie therapy on human colon carcinoma cell transfected with cytosine deaminase gene. Methods: On the basis of SW480 colon carcinoma cell culture, CD suicide gene was transfected into SW480 cell, and then to investigate the effects of SW480 cell cycle and apeptosis by flow cytometer, Results: The decreasing Gt, phase cells, increasing S phase cells were observed significantly in group PDT after 24 hours and 12 hours respectively as compared with that of the control ( P 〈 0, 01 ). The cell cycle distribution of group PDT + CD/5 - Fc has no significantly difference as compared with that of the control( P 〉 0.05 ), But increasing apoptosis ratio was observed in group PDT + CD/5 - Fc as compared with that of the control( P 〈 0.01 ). Conclusion : G1 phase cells maybe more sensitive to PDT than that of S phase cells in our experiment. Gt phase cells and S phase cells maybe both sensitive to PDT + CD/5 - Fc. PDT combination with CD/5 - Fc suicide gene could induce much more apoptosis ratio in SW480-CD cells.
