摘 要:[目的]研究阿魏菇的液体制种技术。[方法]采用一种自制的简易气升式生物反应器研究了阿魏菇液体制种技术,通过正交实验探讨了阿魏菇的最优发酵培养基配比。[结果]不同菌株在不同培养基配方中的菌丝生物量有明显差异,蔗糖(碳源)对菌丝生物量的影响最大,一定量的蛋白胨(氮源)能够促进菌丝生长,少量的K+和Mg2+有利于菌丝生长,但它们对阿魏菇菌丝生长速度影响较小且均未达到显著水平(P〉0.05)。最优培养基为:3.00%蔗糖+0.50%蛋白胨+0.10%KH2PO4+0.05%MgSO4。用该培养基在自制气升式生物反应器中进行阿魏菇液体培养,10 d后菌丝生物量达到25 g/L。空气流速在0.08~0.12 m^3/h有利于菌丝的迅速生长且形成的菌丝球大小均一。栽培实验表明,通过气升式反应器生产的液体菌种可以正常出菇。[结论]阿魏菇液体菌种具有菌丝生长快、制种和生产周期短等优点,是实现阿魏菇规模化、工厂化生产的必然选择。The purpose was to study the liquid strain production technique of Pleurotus nebrodensis.The liquid strain production technique of P.nebrodensis was studied with a simple homemade airlift bioreactor.The optimum fermentation medium of P.nebrodensis was also resarched through orthogonal experiment.The mycelial biomasses of different strains in different medium formulas had obvious difference.The influence of sucrose(carbon source) on mycelial biomass was big;a certain amount of peptone(nitrogen source) could promote mycelial growth,a little of K+ and Mg2+ was in favor of mycelial growth.but their influences on the growing speed of P.nebrodensis mycelial were smaller and didn't reach significant level(P〉0.05).The optimum medium was 3.00% sucrose+0.50% peptone+0.10% KH2PO4+0.05% MgSO4,with which P.nebrodensis was cultured in homemade airlift bioreactor for liquid culture and the mycelial biomass reached 25 g/L after 10 d.The air velocity of 0.08~0.12 m3/h was in favor of the rapid growth of mycelial and the formed mycelial pellets were same in size.The cultivation trial showed that the liquid strain produced by airlift reactor could make P.nebrodensis fruiting normally.The liquid strain of P.nebrodensis had advantages of fast mycelial growth,short strain production and production cycle,which was the necessary choice to realize scale and factory production of P.nebrodensis.
