DNA结合抑制蛋白-1在结肠癌中的表达及其意义的研究

摘  要:目的探讨结肠癌中DNA结合抑制蛋白-1(Id—1)的表达与肿瘤细胞分化程度、浸润深度、淋巴结转移及TNM分期的关系。方法用免疫组化方法分别检测17例结肠腺瘤、16例不典型增生、52例结肠癌及J5例癌旁正常纽织中Id—1的表达。结果正常肠粘膜组织中Id-1蛋白偶有表达,在结肠腺瘤、不典型增生、结肠癌组织中的阳性表达率分别为29.4%、43.75%、73.1%,三者比较差别有统计学意义(P〈0.05)。分化程度低的结肠癌组织中Id.1的阳性表达率高于分化程度高者,两者比较差别有统计学意义(P〈0.05);35例伴有淋巴结转移的肿瘤原发灶者29例(82.9%)表达Id-1,17例无淋巴结转移者9例表达Id-1(52.9%),两者比较有统计学意义(P〈0、05);随着肿瘤浸润深度的增高、TNM分期的升高,Id-1的表达阳性率逐渐升高,差异有统计学意义(P〈0.05)。Id-1蛋白的表达与结肠癌组织的分化程度、浸润程度、淋巴结转移、TNM分期有关,与年龄无关。结论Id-1蛋白在结肠癌组织中表达明显升高,并与结肠癌侵袭转移及TNM临床分期相关,可作为结肠癌预后判断的指标之一。Objective To detect the expression of inhibitors of DNA binding-l(Id-1)protein in human colon cancer and investigate the relationship between the expression of Id-1 protein and histological grade, invasion, lymph node metastasis, TNM stage of colon cancer. Methods Immunohistochemistry was used to detect the expression of Id-1 protein in 52 cases of human colon carcinomas, 15 cases of the adjacent normal colon tissues, 16 cases of dysplasia(DYS), and 17 cases of adenoma. Results The Id-l protein expression was almost not observed in normal colon tissues, and the rate of Id-1 positive expression in adenoma. DYS and colon carcinomas were 29.4%. 43.75%, 73.1%, respectively, which was increased with the lesion progression(P〈 0.05). Id-1 expression in colon carcinomas with low histological grade was higher than that in carcinomas with higher histological(P〈 0.05).Positive rate of Id-1 expression in 35 cases with lymph node metastasis was 82.9%(29/35), which was higher than those without lymph node metastasis(P〈 0.05). With depth of invasion increasing, stage of TNM upgrading, the positive expression of Id-1 protein in colon carcinomas significantly ascended(P〈0.05). The rate of Id-1 positive expression was related to invasion, differentiation, metastasis and TNM stage in colon cancer, and no related to age. Conclusion Id-1 might play an important role in the development of invasion and metastasis of colon cancer.
