LacZ基因作为报道基因在植物基因工程中的应用

摘　　要：通过农杆菌（Agrobacteriumtumfaciens）的介导，将β-半乳糖苷酶基因（Lacz）转入烟草中，得到的转化株能在含有卡那霉素和梭苄青霉素筛选培养基上正常生长，Nopaline检测表明转化体中具有章鱼碱合成酶（NOS）的活性．1．5％的戊二醛可以固定植物中的内源背景，X-gal染色可以很好地显示出外源目的基因LacZ的表达．从而确定广泛用于动物和微生物基因工程的报道基因LacZ可以用于植物基因工程研究．The LacZ gene of Escherichia coli,coding for β -galactosidase, was transferredinto tobacco dises by Agrobacterium tumfaciensmediated These cangrow normally on mediumconsisting of Km. Nopaline synthase activity can be detected in about 95% of these plants.The background of endogenous LacZ activity in tobacco. can be fixed with=1.5%glutaraldehyde, and detectedwith X-gal. From the data .of histochemical stain, It is revealedthat the expression of LacZ gene in transgenic plants, and so LacZ gene as a reporter gene whichis used widely in animal animal and microbe geneengineeringcan be used in plant gene engineering.
