猪皮酸松弛胶原纤维的制备及性能表征

摘　　要：猪皮经酶脱毛、酶和硼氢化钠等前处理,在稀醋酸体系中酸松弛,组织匀浆后,经中和、冷冻干燥制备了酸松弛胶原纤维。对所制得的胶原纤维进行分析测试。结果表明,用该方法制备的酸松弛胶原纤维产率达47.8%,电泳和氨基酸分析结果显示酸松弛胶原纤维纯度高,但有少量的胱氨酸存在。热变性温度为58.2℃,Zeta电位方法测得其等电点为7.44,园二色谱及红外分析结果显示其保持了胶原的天然三股螺旋构象。The acid relaxed collagen fiber was prepared from pigskin, which was unhaired, pretreated with A. S. 1398 protease and sodium borohydride. The pretreated pigskin was relaxed in the dilute acetic acid system, and the relaxed mass was dispersed, neutralized and ice dried to prepare acid relaxed collagen fiber. The purity and properties of the fiber were determined. The results showed that the yield could reach 47.8% on the basis of dry weight. According to the eleetrophoresis and amino acid analysis results, except a little amount cystine existed, the fibers have good purity. The thermal denaturation temperature of the fiber was 58.2 ℃, and its isoelectric point was 7.44, which determined by Zeta potential determination method, and its natural triple helix conformation was maintained according to the circular dichroism and FT-IR results.
