结肠癌细胞中整合素αvβ6-ERK直接连接通路的研究

摘　　要：目的证实结肠癌细胞中整合素αvβ6与细胞外信号调节激酶(ERK)之间存在直接连接。方法应用联合免疫沉淀的方法,观察整合素αvβ免疫耗竭对细胞中ERK的影响,以及ERK免疫耗竭对整合素αvβ6的影响。结果整合素αvβ6和ERK之间存在相互依存的关系,即免疫耗竭整合素αvβ6后,细胞溶液中的ERK明显减少;而免疫耗竭ERK后,细胞溶液中整合素αvβ6也显著降低。结论整合素αvβ6和ERK之间存在着直接的连接,这一直接连接为肿瘤的生长提供了一个重要的信号调节通路。[ Objective ] To search for the direct binding of integrin αvβ6 - ERK in the colon cancer cells. [ Methods ] With the methods of immunoprecipation, studied the alternation of integrin αvβ6 expression after ERK immunodepletion, and the alternation of the ERK binding to 136 after integrin αvβ6 immunodepletion. [ Results] There was an interdependent relationship between integrin αvβ6 and ERK, if one of them was immunodepleted, the other vanished too. [ Conclusion] There is a direct binding between integrin αvβ6 and ERK, which provides an important regulating signal pathway for the growth of colon cancer cells.
