摘 要:目的探讨人脑胶质瘤中EphB4表达情况及临床病理学意义。方法收集正常脑组织和胶质瘤组织,行逆转录-聚合酶链式反应(RT-PCR)和免疫组织化学染色(IHC),分析与病理级别、年龄、性别的相关性。结果EphB4在胶质瘤中呈过表达,表达于肿瘤细胞和血管上,与病理级别正相关,与年龄、性别无明显相关。结论EphB4/ephrinB2信号可能通过直接影响肿瘤细胞和间接影响瘤性血管的双重作用促进肿瘤发生发展。Objective To study the expression of EphB4 in gliomas and its clinicopathological significance. Methods Semi-quantitative RT-PCR and immunohistochemistry were used to examine the expression of EphB4 in normal brain tissue and glioma tissue. Clinicopathological correlations were statistically analyzed in a series of 32 gliomas and 3 nomal brain specimens. Results EphB4 mRNA and protein were overexpessed in the majority of tumor tissues and no expression was observed in nomal brain tissues. In addition, a strong immunohistochemical staining of EphB4 was seen n endothelial cells of tumors. Increased EphB4 mRNA and protein expression were positively correlated with histological grades, but not with gender and age. Conclusion The overexpression of EphB4 is involved in the development of glioma and may play dual effects on tumor and neoangiogenesis.
