摘 要:目的:观察大鼠大脑皮质中代谢型谷氨酸受体5(metabotropic glutamate receptor 5,mGluR5)在脑缺血不同时段的表达及其动态变化的规律,探讨其在急性脑缺血中变化的意义。方法:35只雄性Wistar大鼠随机分为正常对照组、脑缺血1h组、3h组、6h组、12h组、24h组(均为手术组)及其相对应的假手术对照组,采用大鼠大脑中动脉栓塞(MCAO)模型,至上述规定时间点取大脑组织,行HE染色,用光学显微镜观察神经元损伤程度;用免疫组织化学技术判断神经元阳性表达的强弱;用形态分析系统检测各组mGluR5表达的灰度值。结果:各手术组mGluR5表达均高于假手术对照组及正常对照组,与正常对照组相比,缺血1h其表达即有升高,至12h达高峰。而假手术对照组与正常对照组相比无统计学意义。结论:急性脑缺血可引起mGluR5表达上调,提示mGluR5参与了局灶性脑缺血损伤,可能促进了缺血性脑血管病的发展。但结合以前的研究,我们推测其上调也可能是一种为清除谷氨酸(Glu)而作出的反应性增高,是一种维持机体平衡的自我保护机制。Objecive: To observe the expression of mGluR5 and the dynamic regulation in the cortex of rats with focal cerebral ischemia in different phases and to investigate its significance in acute ischemia.Methods:Thirty-five male Wistar rats were raodomly divided into 11 groups: The normal contral,sham operation and ischemic operation(subdivided into 1,3,6,12 and 24-hour) by middle cerebral artery occlusion model.Brain specimens of the rats from each group were obtained for HE staining and immunohistochemistry staining.To observe the damage of neurons with light microscope and to judge the positive expression of neurons,and then to detect the changes of gray scale value in each group.Results:Compared with the normal and each sham control group,the expression of mGluR5 in each ischemia operation group was elevated 1h after ischemia and reached the peak within 12 hours after ischemia.However,there was was no significant difference between normal and sham control groups.Conclusion:Acute cerebral ischemia can cause up-regulated expression of mGluRs,suggesting that up-regulated expression of mGluR5 might be related with focal cerebral ischemia and it contributed to the development of cerebrovascular disease.According to the results of previous study,however,we presume the up-regulated expression might be the responsive increase in order to remove glutamate,which is a protect mechanism for maintain balance of organism.
