摘  要:目的:观察川芎嗪(TMP)对烧伤后大鼠心肌细胞凋亡的影响及初步机制探讨。方法选健康Wistar大鼠50只,随机分为5组,对照组、烧伤组(即B组,造成30%TBSAⅢ度烧伤)、烧伤+TMP组(B+TMP组,烫伤后即刻腹腔注射TMP,20 mg/kg)、烧伤+SB202190组(B+SB组,烫伤后立即注射SB202190,10 mg/kg)及烧伤+TMP+SB202190组(B+TMP+SB组)。放射免疫法测定TNFα含量,末端标记(TUNEL)法检测凋亡细胞、RT-PCR,免疫组化分析心肌Fas、caspase-8、Bcl-2和Bax基因表达,荧光分析法测定Caspase-3活性。结果与对照组比较,B组大鼠心肌细胞凋亡指数(17.46±2.45),心肌中TNFα含量,Fas、caspase-8和Bax蛋白水平及caspase-3活性均显著性升高,但Bcl-2蛋白表达显著低于对照组(P<0.01)。B+TMP组大鼠心肌TNFα含量、心肌细胞凋亡指数(4.15±0.41),心肌Fas、caspase-8和Bax表达水平及caspase-3活性均显著性低于B组,而Bcl-2蛋白表达水平则显著高于B组(P<0.01)。结论TMP对烧伤后心肌细胞凋亡有较好的抑制作用,其机制可能是TMP通过降低TNFα的生成及调节凋亡相关基因的表达。Objictive To investigate the effects of tetramethylpyraxine(TMP)on myocardial apoptosis following severe burns. Methods A total of 50 healthy adult Wistar rats were randomly divided into 5 groups : The control group (n = 10, without burns), the burn group (n= 10, subjected to a 30% TBSA Ⅲ degree scald injury);the TMP treatment group(n=10,treated by intrapleural injection tetramethylpyraxine (20 mg/kg) after scald injury immediately) ; the TMP + SB202190 treatment group(n= 10)treated by intrapleural injection tetramethylpyraxine (20 mg/kg) and SB202190(10 mg/kg), and SB202190 treatment group(n = 10)treated by intrapleural injection SB202190(10 mg/kg). Levels of tumor necrosis factor alpha(TNFα)in plasma and in myocardium were assayed at 18 post burn hours. Terminal deoxynucleotidy transferase-mediatde dUTP fluoescein nick end labeling(TUNEL) and S-P immunohistochemical staining were used respectively to determine the apoptosis and gene expressions of Fas, caspase-8, Bcl-2 and Bax. The caspase-3 activity was detected by Fluorospectrophotometry. Results Apoptosis index(17.46±2.45) ,TNFα contents, levels of fas, caspase-8 ,Bax gene expression and caspase-3 activity in the rat's myocardium of the burn group were all higher than that of the control group,and the expression of Bcl-2 protein in the myocardium of the burn group was significantly lower than that of the control group(P〈0.01) ;In the TMP treatment group,Apoptosis index(4.15±0.41) ,TNFα contents in plasma and myocardium,levels of fas,caspase-8 and bax gene expression and caspase-3 activity in the myocardium were lower than that of the burn group significantly,the expression of Bcl-2 protein in the rat's myocardium was higher than that of the burn group significantly(P〈0.01). Conclusion Tetramethylpyraxine exerts a good antagonist action on the myocardial apoptosis following severe burns,it could inhibit the production of TNFα and regulate the gene expression of fas, caspase-8,
