摘  要:目的观察端粒酶反义寡聚脱氧核苷酸对人胃癌细胞株的作用。方法用端粒酶反义寡聚脱氧核苷酸与人胃癌细胞株共同温育一定时间后,观察细胞形态变化,检测细胞DNA含量的分布,测定端粒酶活性。结果端粒酶反义寡聚脱氧核苷酸能诱导人胃癌细胞凋亡,抑制端粒酶活性,在形态学上表现为细胞膜起泡、染色质固缩、核碎裂、凋亡小体形成;电泳呈凋亡特征性Ladder带;流式细胞仪分析显示,在G1期前出现亚二倍体凋亡峰,端粒酶活性抑制。结论端粒酶反义寡聚脱氧核苷酸能抑制端粒酶活性,诱导胃癌细胞凋亡,从而抑制胃癌细胞的增殖。Objective To observe the effect of telomelase antisense oligodeoxynucleotides on gastric cancer cell line. Methods Gastric cancer cell line was cultured with 1-5μmol/L telomerase antisense ohgodeoxynucleotide for certain time, then the cell activity, morphologic changes and cell-DNA distribution were observed. The apoptotic cells were measured by flow cytometry and telomerase activity were detected by TRAP-ELISA. Results Telomerase antisense ohgodeoxynucleotide could induce the apoptosis of human gastric cancer cell hnes and inhibit the activity of telomerase,which presented apoptotic features: intace cell membrane, chromatin condensation, nucleic fragmentation and apoptotic body formation. Flow cytometry analysis showed that a sub-G1 cell peak and agarose electrophoresis showed a marked DNA ladder, which suggested that telomerase activity was inhibited. Conclusion Telomerase may play an important role iri the growth of human gastric cancer ceils. Telomerase antisense oligodeoxynucleotides may inhibit telomerase activity,which may be a new target of treatment to human gastric cancer.
