摘  要:目的探讨人类重组粒—巨噬细胞集落刺激因子(rhGM-CSF)对辐射物免疫器官的AgNOR定量论功.方法雄性Beagle狗14只,随机分成辐射时抵组4只.高、低剂量rhGM-CSF治疗组各5只.动物均双侧髂骨屏蔽,其它部位用60CO照射.除对照组外.其它两组股静脉注射rhGM-CSF共20d,低剂量组为10μg·kg-1·d-1,高剂量组为20μg·kg-1·d-1,第30d,取出脾脏与淋巴结,组织切片,Ploton’s法AgNOR染色、用分析仪计数100个细胞核内AgNon颗粒数,求其均值。结果低剂量组rhGM-CSF组的AgNOR颗粒数高于对照组(P<0.05),高剂量组rhGM-CSF的AgNOR颗粒数明显高于对照组(P<0.01).结论rhGM—CSF可促进辐射后狗脾及淋巴结形态与功能的恢复,其恢复程度与该因子剂量有关.Objective To explore the effects of rhGM-CSF on the reparatinn of driune organs in irradiated dogs-Angor quantitative analysis.Mathods 14 male Beagle dogs were randornly diveded into 3 groupe, irradintion control group (4 dogs), low and high.dosage rhGM-CSF treat ing groups (5 each group). Afer all dogs were shieded on double illums. they were irradinted by 60Co ray.With the exception of control group, in the other two groups, rhGM-CFS was intwted in femoral vein daily for 20 days 10μg. kg-1.d-1in low dosage group, 20g. kg-1. d-1in high.At the 3othday, the sp1een and lymph nodes were removed out, sectinned and stained with Ploton's Agnor method.Agnor granule number in 100 cells nudei were counted, the average was worked out.Results The Agnor granule number in low dosage rhGM-CSF group is more than that of control group (P<0.05). The Agnor granule number in high dosage rhGM-CSF group is much more than that of control group (P<0.01).Conclusion The rhGM-CSF could promote the recovery of morphosis and function of immune orgasn in irradiated Beagle dogs and that the recovery degree is related to the dosage of rhGM-CSF.
