生姜油对佐剂性关节炎小鼠血清IL-1β和IL-4的影响

摘　　要：目的观察生姜油对佐剂性关节炎(adjuvant arthritis,AA)小鼠血清IL-1β和IL-4的影响。方法将小鼠随机分成5组,即正常对照组、AA模型组、生姜油高剂量组(1.0ml·kg-1)、生姜油低剂量组(0.5ml·kg-1)、地塞米松组(1.0ml·kg-1),然后除正常对照组外,每只小鼠右后足跖皮下注射弗氏完全佐剂(FCA)制备AA小鼠模型。致炎后第8天起开始ig给药,1次/d,连续14d。分别在致炎后第8,12,16,20天测量小鼠足肿胀度,并于第21天处死小鼠,ELISA法测血清中IL-1β和IL-4。结果生姜油1.0,0.5ml·kg-1均能明显减轻小鼠足肿胀(P<0.01或P<0.05),生姜油1.0ml·kg-1可显著降低AA小鼠血清IL-1β(P<0.05),但对IL-4无明显的影响。结论生姜油对AA有抑制作用,其抗炎机制之一可能是降低血清IL-1β水平。Objective To study the effects of ginger oil on serum IL - 1β and IL -4 in AA mice. Methods Mice were randomly divided into five groups ： normal control, adjuvant arthritis （AA） model, high - dose ginger oil （ 1.0 ml·kg^-1 ） ,low - dose ginger oil（0.5 ml·kg^-1 ）and dexamethasone. All mice were injected subcutaneously Freund＇s complete adjuvant （FCA） in right voix pedis , except for normal control. The intragastric administration procedure began at the eighth day after modeling once daily for fourteen days. The changes of paw swelling were measured by volumetric method, Serum IL - 1β and IL -4 were measured by ELISA. Results Ginger oil could significantly inhibit paw swelling in AA mice at the dose of 1.0 - 0.5ml·kg^-1. Ginger oil could remarkably diminish serum IL - 1β at the dose of 1.0 ml·kg^-1, but had no significant effect on serum IL -4. Conclusion Ginger oil has inhibitory effect on AA and the mechanism may be related to dinminish serum IL - 1β.
