摘 要:将NKA-9树脂装柱处理后,应用重度黄疸性疾病新生儿患者的换血血浆分别在静态和灌流状态下进行吸附,分别测定不同时间、不同胆红素浓度下的吸附率。并以吸附柱直径、灌流速度、温度三因素设计三水平正交实验,探索较佳的吸附条件。再测定灌流过程中血浆有效成分的变化量,并做生物相容性实验。结果显示NKA-9树脂对胆红素在静态和灌流状态下的吸附率分别在36%~45%和70%~90%之间。较佳吸附时间为40min、吸附柱直径与体积比为0.20cm^-2、灌流速度为100.56cm/h、温度25℃。在较佳吸附条件下NKA-9树脂对血浆中胆红素的最大吸附率接近95%。实验证明NKA-9树脂对血浆中胆红素的吸附率较高,且血液相容性和生物相容性良好,可作为临床血液灌流法治疗新生儿黄疸性疾病的吸附剂。NKA-9 resin adsorption effect to hematoplasma bilirubin and biocompatibility were investigated. Hematoplasma of serious Icteric disease aegers were perfused by exchanging transfusion in the packed NKA-9 resin under quiet state and perfusion state. Better adsorption time and the number of adsorption columns used to treat Icterus neonatorum disease were ascertained after detecting the adsorption rate at different time and different bilirubinic concentration, respectively. Three level perpendicular experiments were designed under the conditions of the diameter of adsorption column, the flow rate of perfusion and the temperature, better adsorption conditions were explored. Finally the change quantities of the other ingredients of hematoplasma were detected and the biocompatibility experiment was done. The results show that the adsorption rate of NKA-9 resin to bilirubin under quiet state and perfusion state are 36% -45% and 70% - 90%, respectively. The better adsorbing time is 40 min, the ratio of diameter and volume of adsorption column is 0. 20 -2 cm, the flow rate of perfusion is 100.56 cm/h , temperature is 25℃. The adsorption rate of NKA-9 to Bilirubin in hematoplasma is close to 95% under better conditions. NKA-9 resin has high adsorption rate to bilirubin and better blood compatibility and biocompatibility, which indicated that it could be as absorbent for treating Icterus neonatorum disease by clinical hemoperfusion.
