超声导引肝内乙醇注射术后动态观察及再生研究

摘　　要：目的研究在超声导引下经皮乙醇注射(PEI)术后,肝功能恢复及周围肝脏组织是否存在再生及特点。方法取SD大鼠40只,在超声引导下肝内注射无水乙醇0.1ml,分别在第1、3、5、7d取血,检测肝功能,并测定肝组织的有丝分裂指数(MI),增殖细胞核抗原(PCNA),细胞核DNA含量,通过流式细胞仪(FCM)分析S期细胞比率(S-phase cell ratio,SPR)变化。结果PEI术后第1、3d大鼠肝脏MI,SPR及PCNA和细胞核DNA含量均比术前明显增高(P<0.05),并且在第3d达到峰值,第5d再生明显减弱。肝功能在术后第1d升高最明显,随后开始下降,第7d恢复至术前水平。结论PEI术后乙醇局限性扩散所致的肝细胞坏死,可促使其周围肝组织一定程度再生。Objective To study the liver regeneration and its dynamic changes after percutaneous ethanol injection（PEI）into the liver under ultrasonographic guidance.Methods Forty SD rats were performed PEI into the liver under ultrasonographic guidance.The liver function,mitosis index（MI）,proliferation cell nuclei antigen（PCNA）,contents of DNA in cell nuclei and S-phase cell ratio（SPR） of the normal liver tissue were determined prior to the surgery and 1,3,5 and 7 d after surgery.Results Day 1 and 3 after the PEI,the liver tissue MI,PCNA,the contents of DNA in the cell nuclei,and S-phase cell ratio（SPR） were higher than those of the normal （P〈0.05）.At the day 3 after PEI,the liver regeneration was at the top;at day 5,the regeneration was decreased.The liver function was increased most at the first day after PEI.Conclusions The high concentrated ethanol induced by PEI locally diffused to cause the liver cells necrosed,and to enhance the regeneration of the peripheral liver cells.
