摘  要:目的通过对视网膜神经节细胞(retinal ganglion cells,RGCs)微管相关蛋白-1B(microtubule associated protein-1B,MAP-1B)的研究,观察蛇毒神经生长因子(venom nerve growth factor,vNGF)在鼠视神经夹伤后对RGCs的保护作用。方法将Wistar大鼠40只随机分为实验对照组、实验治疗组,每组20只。制作视神经夹伤模型,用头部宽1mm的微型血管夹夹伤大鼠右眼视神经后,实验治疗组向右眼玻璃体内注入vNGF 100 BU(0.025mL)。实验对照组向右眼玻璃体内注入0.025mL平衡盐液。2组的左眼均未做任何处理,作为正常对照组。于损伤后3d、7d、14d、30d、60d取材,在光镜下计数RGCs,透射电镜观察不同时间段各组视网膜形态学变化,并进行MAP-1B免疫组织化学染色分析。结果光镜下,伤后3~30d实验治疗组的RGCs数目高于实验对照组,差异有显著统计学意义(P〈0.01),伤后2组RGCs数目均比正常对照组少,差异有显著统计学意义(P〈0.01)。伤后60d,实验治疗组与实验对照组之间的RGCs数目差异无统计学意义(P〉0.05),实验治疗组、实验对照组与正常对照组差异有显著统计学意义(P〈0.01)。电镜下,伤后14d实验治疗组视网膜微管数目比实验对照组多,排列也较整齐。免疫组织化学结果:实验治疗组2周内MAP-1B表达逐渐增强,1个月时明显减弱;实验对照组3d时有MAP-1B表达,到2周时表达明显减弱。结论在视神经损伤早期,vNGF能减轻视神经夹伤后微管的损坏,提高MAP-1B的表达强度,提高RGCs的存活数量,对RGCs有明显的保护作用。 Objective To study the neuroprotective effects of snake venom nerve growth factor(vNGF) on retinal ganglion cells(RGCs) after optic nerve crush in rat by observing microtubule associated protein-1B(MAP-1B).Methods Forty wistar rats were divided randomly into experimental treatment group and experimental control group,20 in each group.The rat’s optic nerve was crushed with mini-vascular nip.After crushed the optic nerve of the right eyes,100 BU(0.025 mL) of vNGF were injected into the injury eyes of vitreous in experimental treatment group.Balance salt solution(0.025 mL) was injected into the injury eyes of vitreous in experimental control group.The left eyes were not treated with anything as normal control group.In 3 days,7 days,14 days,30 days,60 days after injury,the histological changes of the retina were observed.The numbers of the RGCs were counted by microscope.The morphological changes of the retina were observed by electronic microscope,and the MAP-1B were stained and analyzed with immunohistochemical method.Results Under microscope,in 3~30 days after injury,the number of RGCs in the experimental treatment group was higher than that in the experimental control group(P〈0.01).The numbers of RGCs in the experimental control and experimental treatment groups were lower than normal control group.Their differences were significant (P〈0.01).In 60 days after injury,the differences between the numbers of RGC in the experimental treatment group and experimental control group were not significant(P〉0.05).Under electron microscope,in 14 days after injury,the numbers of microtubules in the experimental group were more than those in the experimental control group obviously.The results of immunohistochemical staining:expression of MAP-1B in the experimental treatment groups was gradually increasing during 2 weeks,and decreasing obviously at one month.At 3 days,it was showed that there were expression of MAP-1B in the experimental control groups and decreasing obviously at t
