摘 要:目的研究金雀黄素抑制人乳腺癌细胞株体外增殖作用及其作用机理。方法选用两种不同的人乳腺癌细胞株MCF-7和MDA-MB-231体外培养,MTT法检测细胞增殖作用,Giemsa染色观察细胞形态学改变,流式细胞仪检测细胞周期及凋亡率,观察不同剂量的金雀黄素诱导细胞凋亡。同时采用免疫组化检测凋亡相关基因Bax和癌基因erbB-2蛋白的表达。结果金雀黄素对MCF-7细胞和MDA-MB-231细胞体外生长具有明显的抑制作用。Giemsa染色显示,金雀黄素处理的MCF-7细胞呈明显的凋亡形态改变,细胞固缩、发泡,染色质凝集呈块状,并沿核周分布。流式细胞仪检测在G1峰前可见凋亡峰,且随用药时间的延长,凋亡比率递增。而金雀黄素处理的MDA-MB-231细胞未见明显的凋亡形态改变和凋亡峰的出现,细胞周期明显地阻滞于G2-M期。蛋白水平检测表明金雀黄素促进MCF-7细胞Bax表达升高,抑制erbB-2蛋白的表达。结论金雀黄素通过诱导细胞凋亡或阻滞细胞于G2-M期,从而抑制乳腺癌细胞的体外增殖,并主要是通过调节Pax和erbB-2蛋白的表达实现的,为其减缓人乳腺癌细胞的体内生长提供理论依据。Objective To study the mechanisms of inhibiting proliferous effect of human breast cancer call line treated with Genistein in vitro. Methods Two human breast cancer cell lines (MCF - 7 and MDA - MB- 231 ) were cultured in vitro, proliferation of them was measured with MTT method, cell morphology was observed with Giemsastain, cell cycle and apoptosisrate were measured with flow cytometry and apoptosis was detected in situ. Results Genistein markedly inhibited proliferation of MCF- 7 and MDA - MB - 231 cell line. Typical image of apoptosis was observed in MCF - 7 cell line treated with Genistein in Giemsastain: cell eondensation, blebs formation on cell surface and pieces of condensed chromatin were scattered along nuclear envelope. The results detected with flow cytometry showed GI subpeak before G1 phase peak (apoptosis peak). Apoptosis rate increased withtime of Genistein treatment. However, MDA- MB- 231 cell line treated with Genistein did not show apoptosis and apoptosis peak, and quantity of cells at G2- Mphase obviously increased. Conclusion Genistein inhibites proliferation of human breast cancer cellline in vitro through inducing apoptosis or arresting cell at G- M phase. Genistein can induce MCF27 cells apoptosis and it may be one of the mechanisms for theinhibitory effect of genistein in human cancer cells.
