摘 要:目的探讨洛铂体外诱导人卵巢癌顺铂耐药SKOV3/DDP细胞凋亡及其机制。方法采用四甲基偶氮唑盐(MTT)法测定不同浓度和时间洛铂对SKOV3/DDP细胞的生长抑制作用,并与其亲本细胞SKOV3(敏感株)相比较;进一步通过光镜及透射电镜观察SKOV3/DDP细胞的形态学改变;并采用流式细胞仪检测细胞周期变化。结果洛铂作用SKOV3/DDP细胞的增殖呈时间剂量依赖模式受到抑制,与SKOV3细胞相比,差异无统计学意义(P>0.05);一定浓度的SKOV3/DDP作用一定时间,可诱导SKOV3/DDP细胞发生凋亡;光镜和透射电镜下可见典型的凋亡细胞形态学改变;流式细胞技术分析发现洛铂作用首先引起细胞周期分布的改变,随用药时间的延长,凋亡率逐渐升高。结论洛铂通过诱导凋亡而抑制体外培养的SKOV3/DDP细胞生长,其机制可能与细胞G0/G1期阻滞有关。Objective To investigate the apoptosis-inducing activity of Lobaplatin(Lob) on cisplatin-resisrant human ovarian carcinoma cell line SKOV3/DDP and its related mechanism. Methods Cell proliferation in different concentration and time was determined by MTT (Methyl thiazolyl tetrazolium) assay, and compared with other parent cell SKOV3. Then light microscope and transmission electron microscope were used to examine the morphologic changes of SKOV3/DDP cells. And cell cycles and apoptosis were examined by flow cytometry (FCM). Results The result of MTT colormetric assay showed that proliferation of SKOV3/DDP cells was inhibited by lobaplatin in a dose-time-dependent fashion, and there was no significant difference (P〉0. 05)in two cell lines. When SKOV3/DDP cells were treated with lobaplatin at a certain concentration,the morphologic changes were found by light, transmission electron microscope and flow cytometric analysis. Flow cytometric analysis showed that after using lobaplatin, cell cycle distribution changed at first and as the time prolonged apoptotic peak went higher. Conclusion Lobaplatin inhibits proliferation of SKOV3/DDP cells by inducing apoptosis and G0/G1 phase arrest may be involved in the mechanism.
