摘　　要：目的探讨血管内皮生长因子(VEGF)在羊膜为载体培养的角膜上皮细胞中的表达情况。方法体外羊膜为载体培养大鼠角膜上皮细胞(羊膜载体组),以无载体培养的角膜上皮细胞为对照组。逆转录聚合酶链反应(RT-PCR)检测两组细胞VEGF mRNA表达,酶联免疫吸附试验(ELISA)检测两组细胞VEGF蛋白表达。结果羊膜载体组角膜上皮细胞VEGF mRNA含量为0.66±0.08,蛋白表达量为1 605.22±196.08;对照组角膜上皮细胞VEGF mRNA含量为0.83±0.10,蛋白表达量为1 614.39±142.43;两组细胞VEGF表达水平无显著差异。结论羊膜载体不能有效抑制角膜上皮细胞VEGF的表达。Objective To explore the expression of vascular endothelial growth factor （VEGF） in cultured corneal epithelial cells with amniotic membrane. Methods Rat corneal epithelial cells were cultured in vitro with amniotic membrane （ amniotic membrane group） , and those cultured without amniotic membrane were served as control group. The expression of VEGF mRNA in the cells of the two groups was examined by reverse transcriptase-polymerase chain reaction （RT-PCR） , and the expression of VEGF protein was determined by enzyme-linked immunosorbent assay （ELISA）, Results The expression level of VEGF mRNA of the amniotic membrane group was 0.66 ± 0.08, while that of the control group was 0.83 ±0, 10, The expression level of VEGF protein of the amniotic membrane group was 1 605.22 ± 196.08, while that of the control group was 1 614.39 ±142.43, There was no significant difference in the expression level of VEGF between the two groups. Conclusion Amniotic membrane can not effectively inhibit VEGF expression of cultivated corneal epithelial cells.
