癫痫发作对大鼠学习的影响及其与铜、锌含量的关系

摘　　要：目的探讨癫痫发作对大鼠学习记忆过程的影响及其与微量元素铜、锌的关系。方法以雄性听源性癫痫易感大鼠５０只为实验对象，随机均等分为５组，以１００ｄＢ铃声刺激诱发癫痫，以光分辨〔三等分辐射式迷宫箱内，光信号示安全区，大鼠逃脱足底电击进入安全区为正确反应〕作为学习记忆指标，分别处理动物。记录每只大鼠的学习成绩及达标所需的天数，末次实验结束后，处死动物，取出所需组织，用原子吸收分光光度法测定组织中铜、锌的含量。结果癫痫对大鼠学习记忆的不同阶段均有影响，频发癫痫严重干扰学习过程，并且伴有颞叶皮质、海马、下丘脑及肾上腺组织内铜、锌含量改变。结论癫痫可影响学习记忆过程，其机理可能与脑内依赖铜、锌的酶的活性改变有关。Objectve To study the effects of epilepsy on learning and Memory and the contents of Cu 2+ and Zn 2+ in temporal lobe cortex,hippocampal gyrus,hypothalamus and adrenal gland.Methods 50 audiogenic epilepsy seneitive rats were randomly divided 5 groups:C: control group;M+E:first learning then inducing epilepsy group;E+M:first inducing epilepsy then learning group;E6+M:first inducing frequent epilepsy then learning group.After the last experiment each rat was sacrificed and the tissues to be tested were taken out.The contents of Cu 2+ and Zn 2+ were measured by the method of atomic absorption.Results The results showed epilepsy could interrupt whole learning process(including importation,maintenance and extraction of message).Compared with control group the content of Cu 2+ during learning process were decreased in hippocampal gyrus,hypothalamus and cortex and medulla of adrenal gland,but no obvious differece in temporal lobe cortx.The content of Zn 2+ were increased in cortex and medulla of adrenal gland and decreased in temporal lobe cortex and hypothalamus but there was no obvious change in hippocampal gyrus. Frequent epilepsy could markly enhence the content of Zn 2+ and decrease the content of Cu 2+ in all tested tissues.Conclusion learning and epilepgy interrupting learning processes are related to the changes of Zn 2+ in temporal lobe cortex and Cu 2+ in hippocampal gyrus and both could cause the changes of metabolism of endocrine glands.
