基质金属蛋白酶-9、血管内皮生长因子基因在胰腺癌中的表达及其临床意义

摘　　要：目的研究基质金属蛋白酶-9(MMP-9)、血管内皮生长因子(VEGF)基因在胰腺癌中的表达,探讨它们与胰腺癌预后的关系。方法用逆转录-聚合酶链式反应(RT-PCR)技术检测35例胰腺癌(包括癌旁组织及正常组织)手术标本中VEGF和MMP-9 mRNA的表达。结果MMP-9、VEGF在胰腺癌组织中的表达水平显著高于癌旁组织及正常组织,且其表达水平与胰腺癌的分化程度有关,且在淋巴结转移组与非转移组间其表达水平的差异有统计学意义(P<0.05)。结论MMP-9、VEGF基因可作为反映胰腺癌侵袭转移、判断预后的生物学指标。Objective To evaluate the expression of matrix metallo proteinases-9 （MMP-9）, vascular endothelial growth factor （VEGF） and their correlation in pancreatic carainoma. Methods VEGFmRNA and MMP-gmRNA were examined by reverse transcription-polymerase chain reaction （RT-PCR） in 35 PC tissue. Results The expression of MMP-9, VEGF was higher in PC than that in its surrounding parenchyma and normal pancreatic parenchyma. The expression of MMP-9, VEGF were related to the tumor differentiation and the lymphnode metastasis. Conclusion The expression of MMP-9, VEGF in PC was higher than that in the parenchyrna surrounding the cancer and the normal pancreatic parenchyma. High level of both VEGF and MMP-9 expression is a predictor for poor prognosis of PC.
