摘 要:目的探讨CD137(4-1BB)激发型单克隆抗体2A对小鼠未成熟树突状细胞株DC2.4表面TLR4表达的调节。方法以流式细胞术检测不同剂量2A、LPS,或LPS预刺激后再加入2A等因素作用下,DC2.4表面的TLR4表达情况。结果LPS下调TLR4表达,2A使DC2.4上TLR4表达上调,并拮抗LPS对TLR4的下调作用。LPS预处理后再加入2A,也拮抗LPS的下调作用。结论CD137信号可以上调DC2.4表面TLR4的表达。Objective To study the regulatory effect of agonist CD137 (4-1BB) monoclonal antibody 2A on TLR4 expression in a routine immature dendritic cell line DC2.4. Methods Flow cytometry was used to investigate the TLR4 expression on DC2.4 under following conditions: different doses of 2A, LPS or addition of 2A after prestimulation of DC2.4 by LPS. Results LPS could down-regulate TLR4 expression on DC2.4. On the contrary, 2A could up-regulate TLR4 expression on DC2.4. Moreover, 2A could inhibit the down-regulatory effect of LPS on TLR4 expression. When DC2.4 was stimulated by 2A after LPS prestimulation, 2A could still up-regulate TLR4 expression. Conclusion CD137 signal could up-regulate the expression of TLR4 on dendritic cell line DC2.4.
