乳腺癌中Survivin的表达及其与Caspase-3和临床病理学特征的关系

摘　　要：目的研究乳腺癌中Survivin的表达及其与Caspase-3和临床病理学特征的关系。方法用免疫组化S-P法检测乳腺癌石蜡标本中Survivin和Caspase-3的表达,并分析Survivin与临床病理学特征的关系。结果①Survivin和Caspase-3蛋白在乳腺癌阳性表达率分别为67.8%和41.4%,两者的表达呈负相关性(rs=-0.720,P(0.05);②Sur-vivin与淋巴结转移和肿块大小有关,与年龄、组织学分级、临床分期及Cerb-B-2表达无关。结论在乳腺癌组织中,Sur-vivin可能通过抑制Caspase-3的活性来抑制凋亡,促使肿瘤增殖。Survivin在乳腺癌中高表达,且与淋巴结转移和肿块大小有关。检测Survivin的表达也许可以预测恶性程度和预后。Survivin有望成为乳腺癌治疗的新靶点。Objective To study the expression of Survivin in breast cancer and its relationship with Caspase-3 and clinicopathological characteristics. Methods The expression of Survivin and Caspase-3 in breast cancer paraffin embedded specimens was detected by Immunohistochemical technique （SP）. The relationship between Survivin and clinicopathological characteristics was analyzed. Results ~The rate of positive expression of Survivin and Caspase-3 protein in breast cancer was 67.8% and 41.4% respectively, and the correlation was negative （ rs = - 0. 720, P 〈 0.05 ） ; ② There was significant correlation between Survivin expression and tumor size and lymph node involvement （ P 〈 0.05 ） ,but not obvious with other clinicopathological factors,such as age, histological grade, stage grouping and Cerb-B-2. Conclusions Survivin may contribute to the proliferation of breast cancer cells through inhibit Caspase-3. High expression of Survivin can be found in patients with breast cancer and it is significantly correlated with lymphoid metastasis and tumor size. Detection of the expression of Survivin may predict malignant degree and prognosis. Survivin may be a new therapeutic target in cancer therapy.
