摘 要:目的:探讨超声微泡造影剂促进GFP质粒转染卵巢癌(SKOV3)细胞的有效性。方法:将SKOV3细胞分为3组:第1组为脂质体组,加入脂质体和GFP质粒;第2组为超声单独作用转化组,加入脂质体、GFP质粒并以一定能量的超声分别辐照30s、60s和5min;第3组为超声介导造影剂组,加入脂质体、质粒和微泡,并用相同能量的超声分别照射30s、60s、5min,每一时间脂质体浓度分别采用2、5和10μl/ml。转染24—48h后用荧光显微镜、流式细胞仪评价GFP转染率。结果:用超声触发破坏微泡促进GFP质粒转染SKOV3细胞,其转染效率明显高于其他两组;其中脂质体5μl/ml超声照射60s组转染效率最高。结论:利用低频率和一定能量的超声击碎微泡造影剂能有效地提高GFP质粒在SKOV3细胞中的转染效率。Objective:To investigate whether ultrasound microbubble intensifier could effectively enhance the transfection efficiency of GFP plasmid into the ovarian carcinoma( SKOV3 ) cells. Methods:SKOV3 cells were divided into three groups. The first group was given proper dose of liposome and GFP plasmid. The second group was exposed to ultrasound for 30seconds, 60seconds and 5minutes with the same dose of liposome and GFP plasmid. The third group was also exposed to ultrasound for the same time with liposome, GFP plasmid and microbubbles, and liposome in the time of each group was selected 2μl/ml,5μl/ml and 10μl/ml. The expression of GFP in SKOV3 cells was examined by flow cytometry and fluorescence microscopy. Results: The transfection efficiency of the third group was significantly higher than that of the other two groups. When liposome was given at dosage of 5μl/ml and exposed to ultrosound for 60s, the transfection efficiency was the highest. Conchlsion:Ultrosound-mediated microbubbles destruction can significantly enhance the transfection efficiency of GFP in SKOV3 cells.
