植酸酶毕赤氏酵母诱导条件初步研究

摘　　要：目的优化产植酸酶的毕赤酵母基因工程菌的表达条件,确定最佳的诱导表达条件。方法先培养酵母细胞,然后进行诱导产酶,最后进行酶活测定。结果植酸酶的最佳收获时间为3 d;甲醇的最佳诱导浓度为10 g/L。结论在诱导开始之后加入0.4%的PTM1和0.1%酪蛋白水解物可以明显促进植酸酶的表达。Objective： To optimize phytase expression condition of the recombinant Pichia pastoris and determin expression condition. Methods： Pichia pastoris cells were cultivated firstly and then phytase was induced,finally phytase activity was measured. Results： Optimal harvest time for phytase is 3d and optimal methanol inducement concentration is 10g/L. Conclusion ： 0.4% PTM1 and 0. 1% casamino acid can accelerate the phytase expression obviously after inducement is initiated.
